Different expression patterns of sperm motility-related genes in testis of diploid and tetraploid cyprinid fish†

Abstract: Sperm motility is an important standard to measure the fertility of male. In our previous study, we found that the diploid spermatozoa from allotetraploid hybrid (4nAT) had longer durations of rapid and slow progressive motility than haploid spermatozoa from common carp (COC). In this study, to explore sperm motility-related molecular mechanisms, we compared the testis tissues transcriptomes from 2-year-old male COC and 4nAT. The RNA-seq data revealed that 2985 genes were differentially expressed between COC a… Show more

“…At present, RNA-Seq has been widely used in fish genetic studies (Xu et al 2015; Hu et al 2017; Tao et al 2013; Bar et al 2016; Sharma et al 2014). However, the differences in transcript abundance and the presence of different isoforms greatly challenge the assembly of a transcriptome from short reads.…”

“…Our findings showed a significantly positive correlation of DEG analysis results between RNA-seq and qRT-PCR, particularly for DEGs that had a high log 2 fold change. Similarly, several investigations of transcriptomic analysis have described a positive correlation between RNA-seq and qRT-PCR data 11,59,60 . With both methods of gene expression analysis, the differential expression of several genes that play an important role in regulating reproductive functions were similar to previous transcriptomic analysis findings.…”

“…DExD-box helicase 39B ( DDX39B ) and poly(A) binding protein cytoplasmic 1 ( PABPC1 ) are involved in RNA metabolism ( Ozturk et al, 2012 ; Xu et al, 2014 ; Raj et al, 2019 ). CD74 molecule ( CD74 ) controls the antigen presentation for immune responses and reported in the testes of chickens ( Singh et al, 2015 ) and cyprinid fish ( Hu et al, 2017 ).…”
