Diethyldithiocarbamate-Induced Cytotoxicity and Apoptosis in Leukemia Cell Lines

Abstract: Diethyldithiocarbamate (DDTC) has been shown to induce cytotoxicity in several different systems. We examined whether the DDTC-induced cytotoxicity was via apoptosis, or in relation to intracellular glutathione (GSH) in various murine and human leukemia cell lines. The cells most sensitive to DDTC-induced cytotoxicity were P388 lymphoid neoplasma cells and NALM-6, a B cell line of acute lymphocytic leukemia (ALL). The next level of susceptible cells included J774.1, having a macrophage function, HL-60 premyelo… Show more

“…All cells were routinely kept in RPMI 1640 medium supplemented with 10% fetal bovine serum and penicillin G (100 U/ml)/streptomycin (100 mg/ml) at 37°C in a humidified 5% CO 2 -95% air incubator under standard conditions. 9) Cell viability was measured by counting, excluding those cells stained with 0.2% trypan blue. To maintain exponential growth, cells were seeded at 1ϫ10 5 cells/ml and passaged every 4 to 5 d.…”

“…Cells were cultured in RPMI 1640 medium (Gibco Invitrogen Co., Grand Island, NY, U.S.A.) supplemented with 10% FBS, 100 U/ml of penicillin, and streptomycin (Sigma, U.S.A.) at 37°C in a humidified 5% CO 2 -95% air incubator under standard conditions. 6) To maintain exponential growth, cells were seeded at 1ϫ10 5 cells/ml and passaged every 4 to 5 d.…”

“…K562 has a higher intracellular total glutathione content and lower susceptibly to H 2 O 2 than these other cell lines, whereas Jurkat cells have a lower intracellular total glutathione content than the other human cell lines tested (29). Thus, cells with a lower concentration of intracellular glutathione may be more sensitive to acacetin.…”
