Market instability, increased competition, escalating price and reduced availability of conventional ingredients warrants the researchers to rely on alternative feed ingredients. This approach may help in producing aqua feeds in a sustainable and cost-effective way to accomplish the global food and nutritional securities. Mahua oil cake (Bassia latifolia) is an underutilized non-conventional ingredient that holds promise for incorporation into aqua feed following nutrient enhancement by solid-state fermentation. A five-month pond feeding trial was carried out to investigate the effects of Sachharomyces cerevisiae and Bacillus subtilis fermented mahua oil cake (MOC) on the production performance, nutrient utilization, digestive capacity, and innate immunological responses of Labeo rohita fingerlings. For this, two iso-nitrogenous feed were formulated and prepared incorporating fermented MOC at different levels i.e. 0 and 40 percentage replacing soybean meal and other feed ingredients and fed to rohu fingerlings of two treatment groups in pond culture for 5 months duration. Improved growth performance, feed conversion ratio, feed intake, protein efficiency ratio and digestive capacity were observed in fish fed diets with 40 percent of fermented MOC compared to control. Innate immune responses parameters (respiratory burst activity, myeloperoxidase, lysozyme and hemaglutination activities) were significantly higher (P < 0.05) in fishes fed with fermented MOC. Therefore, we conclude incorporation of solid state fermented mahua oil cake up to 40% level in diet of L. rohita fingerlings in pond culture without any adverse effects on growth, nutrient utilization and innate immune response.

Background: With remarkable growth in aquaculture, the need for system diversification and species diversification has been demanded by the producers to gain fruitful output in terms of profit. Striped murrel, Channa striata has been identified as a suitable species for modern aquaculture systems in terms of species diversification due to their fast growth rate, air-breathing nature, higher market price and consumer acceptance. The major bottlenecks in seed production of murrel are captive maturation and short spawning window. To overcome these problems, an attempt was made to achieve early gonadal maturity of striped murrel in captive conditions by intervening in the brooder rearing habitat. Methods: In this regard, four habitats were tested in duplicates, namely T1 - Tank with clean reservoir water without macrophytes and soil base, T2 - Tanks provided with floating (Water hyacinth, Eichhornia sp) and submerged (Waterthyme, Hydrilla sp) aquatic macrophytes without any soil base, T3- Tanks provided with only soil base (15-20 cm) without any macrophytes and T4 - Tanks provided with both soil base and macrophytes. Result: At the end of the three-months rearing phase, the fishes provided with macrophytes and soil base (T4) resulted in early maturation characterized by an egg size of around 1.29±0.21 mm and the majority of late granular stage oocytes and spermatozoa in ovary and testes, respectively. The results concluded that providing a bottom soil base of 15-20 cm and covering the 20-30% water spread area using aquatic macrophytes in the brood rearing tank helped in the gonadal maturation of striped murrel during pre-monsoon in captive conditions.

An analysis of the extent of the effect of steroidal saponin addition on glucose and lipid metabolism in hybrid grouper liver was performed at the transcriptomic and metabolomic levels. Feeds (52% crude protein, 14% crude lipid) were prepared containing 0% (S0), 0.1% (S0.1), and 0.2% (S0.2) steroidal saponins. After eight weeks of feeding trial, compared to the S0 group, the activities of serum albumin, alanine aminotransferase, and aspartate transaminase were significantly lower and the activities of lysozyme, acid phosphatase, and alkaline phosphatase were significantly higher in the S0.1 group (p < 0.05). The superoxide dismutase, catalase, and glutathione peroxidase activities in the livers of the S0.1 group were significantly higher than those of the S0 group, while the malondialdehyde content was significantly lower than that of the S0 group (p < 0.05). There were forty-two differentially expressed genes and thirty-two differential metabolites associated with glucose and lipid metabolism enriched using KEGG and GO. In the S0 group, the expression of prostaglandin-endoperoxide synthase 1, prostaglandin E synthase 1, and thromboxane-2 synthase mRNA was significantly higher than in the S0.1 group (p < 0.05). The expression levels of genes in the S0 group were significantly higher than those in the S0.1 group (p < 0.05), including for glycogen synthase kinase, glucose-6-phosphatase catalytic subunit 2, fructose-1,6-bisphosphatase, phosphoenolpyruvate carboxykinase, glucose transporter 4, and malate dehydrogenase. The expression of mRNA such as fatty acid synthase, acetyl-CoA carboxylase, and sterol regulatory element-binding protein 1 was significantly lower in the S0.1 group than in the S0 group, while the expression of carnitine acyltransferase 1, acyl-CoA synthetase, and acyl-CoA dehydrogenase genes was significantly higher in the S0 group (p < 0.05). In summary, glycogen synthesis, gluconeogenesis, and the arachidonic acid metabolism pathway were inhibited by 0.1% steroidal saponins, and glycogenolysis, glycolysis, the tricarboxylic acid cycle, and the fatty acid β-oxidation pathway were activated. This study aims to provide a reference for the formulation of grouper feeds with a higher crude-lipid level.
