Dielectrophoresis of malignant human melanocytes

Abstract: Dielectrophoretic experiments on pearl-chain formation and collection of human melanocytes were performed as a function of the frequency of the applied nonuniform electric field. Evidence is reported showing that the behavior of malignant melanocytes is markedly dependent upon the type of established cell line (pigmented or achromic), age, and drug treatment (e.g., chlorpromazine).

“…80 Aer this, DEP equipment has been mainly used in the enrichment and isolation of yeast cells. Since 1982, when human malignant melanocytes were rst successfully arranged by DEP, 81 DEP has been widely used in the manipulation of tumor cells. In 1984, the blood cells obtained from patients suffering from sickle cell anemia were rst used to distinguish normal red blood cells 85 Pd, 86 CdTe, 87 Silica, 88 PMMA, 72 and sickle cells by DEP.…”

“…Early DEP characterizations were based on observations of cell behavior under an applied field. For example, malignant myocytes were characterized by their pearl‐chaining ability and mean chain length as a function of frequency [66]. Dielectric coefficients, measured from the dielectrophoretic velocities of single cells characterized myeloma, hybridomas, and B‐lymphocytes based on their type, age, and drug treatment as well as chloroplasts untreated and treated with cationized ferritin demonstrating differences in surface charges [67,68].…”

“…Human skin cancer cells have specific electrical properties that facilitate separation when using DEP. One important aspect revealed since the earliest experiments [109], is the role of melanin content in modulating melanoma cells' behavior in a non-uniform electrical field. Modifications in cell structure due to normal ageing or destructive changes under different drug therapies will alter the cell movement under DEP, facilitating or impeding single cell separation.…”
