DICER governs characteristics of glioma stem cells and the resulting tumors in xenograft mouse models of glioblastoma

Abstract: The RNAse III endonuclease DICER is a key regulator of microRNA (miRNA) biogenesis and is frequently decreased in a variety of malignancies. We characterized the role of DICER in glioblastoma (GB), specifically demonstrating its effects on the ability of glioma stem-like cells (GSCs) to form tumors in a mouse model of GB. DICER silencing in GSCs reduced their stem cell characteristics, while tumors arising from these cells were more aggressive, larger in volume, and displayed a higher proliferation index and l… Show more

“…It is known that the loss or insufficient expression of Drosha, Dicer, and TRBP can lead to the development of a tumor process (Olejniczak et al, 2018). Several groups showed impaired expression of Drosha and Dicer in glioblastoma, pineoblastoma, and neuroblastoma, all of which correlated with a poor prognosis of survival (Lin et al, 2010;Mansouri et al, 2016;de Kock et al, 2020). Decreased expression of these proteins can be mediated by mutations or epigenetic inactivation of their genes.…”

“…GSC 8-11: Glioma sphere-forming cells (GSC) 8-11 were isolated from surgical sample from a female patient, following written consent and approval by the institutional review board of The University of Texas M. D. Anderson Cancer Center, and have been characterized in the literature [33]. As reported in the literature [34,35], cells were grown in Neurosphere Media containing DMEM/F12 (Corning, Corning, NY, USA) with B27 (×1, Thermo Fisher Scientific, Waltham, MA, USA), bFGF (20 ng/mL, Millipore Sigma, St.…”
