The Role of MicroRNAs in Therapeutic Resistance of Malignant Primary Brain Tumors

Abstract: Brain tumors in children and adults are challenging tumors to treat. Malignant primary brain tumors (MPBTs) such as glioblastoma have very poor outcomes, emphasizing the need to better understand their pathogenesis. Developing novel strategies to slow down or even stop the growth of brain tumors remains one of the major clinical challenges. Modern treatment strategies for MPBTs are based on open surgery, chemotherapy, and radiation therapy. However, none of these treatments, alone or in combination, are consid… Show more

“…MicroRNAs (miRNAs) are small non-coding RNAs consisting of 18-20 nucleotides that play an important role in the regulation of gene expression at the post-transcriptional level by interacting with 3’-untranslated regions (3’-UTR) of messenger RNAs (mRNA)-targets ( 5 ). MiRNAs are involved in the regulation of such physiological processes as cell proliferation, differentiation, apoptosis, angiogenesis, etc.…”

“…MiRNAs are involved in the regulation of such physiological processes as cell proliferation, differentiation, apoptosis, angiogenesis, etc. MiRNAs play an important role in the regulation of both physiological processes ( 5 ). On the other hand, miRNAs have been found to be involved in the oncogenesis of many human tumors, acting as tumor suppressors or oncogenes ( 6 ).…”

“…A novel BET inhibitor, UM-002, reduced the cell proliferation in patient-derived xenograft GBM cell lines GBM22 and GBM39 [112]. MicroRNAs are non-coding RNAs that regulate gene expression of cell cycle regulatory pathways [96]. Downregulation of microRNA-21 induces in GBM cell lines a G0/G1 cell cycle arrest and increased apoptosis and inhibits chemotherapeutic resistance to doxorubicin [114].…”

“…By debilitating the cancer cell’s ability to function optimally for survival and tumorigenesis, chemotherapeutic agents eventually result in the death of cancer cells. Often, chemotherapy is employed in adjuvant settings along with surgery, radiation therapy and even with other chemotherapeutic drugs [ 2 ]. Despite the many benefits of chemotherapy for cancer cure, the associated toxicity is also well known [ 3 ].…”
