Bovine tuberculosis (bTB) is a zoonosis caused by Mycobacterium bovis, a species belonging to the Mycobacterium tuberculosis complex (MTC) group. Direct bTB diagnosis from suggestive lesions can be performed by nested q-PCR targeting the Rv2807 gene present in the MTC group, as well as the TbD1 gene, present in M. bovis. In this context, the aim of the present study was to assess the importance of considering positive MTC results for the Rv2807 target gene obtained through the nested real time polymerase chain reaction (nested q-PCR) applied to samples obtained directly from suspected bTB lesions. A total of 174 samples of suggestive bTB caseous lesions were obtained during cattle slaughter in slaughterhouses in the state of Mato Grosso, Brazil. DNA was extracted from the lesions and nested q-PCR was performed to detect both MTC and M. bovis. Both samples positive for the Rv2807 (41/174) and TbD1 (29/174) were submitted to bacterial culturing (23/41), and the DNA of the isolates (23) was extracted and submitted again to nested q-PCR. The Rv2807 gene (MTC) was previously amplified by nested q-PCR directly from the lesions, although the TbD1 gene specific for M. bovis was not amplified previously in four of the successfully isolated samples (4/23), only following isolation, and only the Rv2807 gene was amplified before and after isolation. In conclusion, the target gene Rv2807(MTC) exhibited higher positivity in the analyzed samples compared to the TbD1 gene (M. bovis).

Between November 2017 and August 2018, in order to determine the occurrence of tuberculosis in cattle slaughtered in the semi-arid region of Rio Grande do Norte, 11,610 bovine carcasses underwent a routine post-mortem inspection. One animal presented suggestive lesions and samples from lung, spleen and heart were submitted to microbiological culture in Stonebrink medium for 90 days and molecular analysis by nested-PCR. For histopathological examination and Ziehl Neelsen staining, the omentum was used. In the cultured samples, two of them (heart and spleen) showed growth, but were not confirmed as M. bovis by conventional PCR. At nested-PCR, the samples showed amplification for the TbD1 region. The presence of numerous granulomas was detected in the histopathological examination characterized by a necrotic center and areas of mineralization, as well as the presence of acid-fast bacilli (AFB) in the Ziehl-Neelsen stain. Microbiological culture can show false negative results, despite being considered a gold standard technique, although it takes time. Nested-PCR and histopathology show fast and effective results for the diagnosis of the disease. The presence of positive animals represents a public health risk in the studied region. Therefore, one of the essential systems applied to the control of bovine tuberculosis is the epidemiological surveillance of animals in slaughterhouses.

Pathological changes in palatine tonsils of slaughtered water buffaloes (Bubalus bubalis)

This study evaluated palatine tonsils from 327 water buffaloes collected in a slaughterhouse located in the city of Belém, Pará State, Brazil, over four months (nine sample collecting visits). The overwhelming majority (84.71%) of these buffaloes came from Marajó Island, Pará State, Brazil. The main pathological findings were crypt dilatations/keratin cysts (96.0%, 314 cases), mild to moderate follicular lymphoid hyperplasia (93.0%, 304 cases), crypt abscesses (79.8%, 261 cases), tonsillar corpuscles (31.2%, 102 cases) and tuberculoid granulomas (1.53%, 5 cases). Acid-fast bacilli (AFB) were observed in 20% (1 out of 5 cases) of these granulomas. The study indicates that pathological evaluation of tonsils in buffaloes can provide details of structural features of the organ in the species and information on subclinical or asymptomatic tonsillitis.
