Diagnostic Values of miR-221-3p in Serum and Cerebrospinal Fluid for Post-Stroke Depression and Analysis of Risk Factors

Abstract: Background: We aimed to explore the diagnostic values of miR-221-3p in serum and cerebrospinal fluid (CSF) for post-stroke depression (PSD) and to analyze the risk factors of the disease. Methods: Admitted to the Second Affiliated Hospital of Harbin Medical University, Harbin, China from May 2013 to May 2020, 136 stroke patients were enrolled, among which 76 PSD patients were taken as a PSD group and 60 non-depressed patients were taken as a Non-PSD group. miR-221-3p expression in serum and CSF and conce… Show more

“…Accordingly, our results revealed that m6A level was decreased as a response to METTL3 downregulation, and miR‐221‐3p in hippocampal tissues showed the same trend of METTL3, while pri‐miR‐221 showed an opposite trend of METTL3. miR‐221‐3p was previously found to be upregulated in the serum and cerebrospinal fluid of DD patients, and increased miR‐221‐3p indicates a high risk of DD (Cui et al, 2021; Enatescu et al, 2016; Feng et al, 2019). To analyze the role of miR‐221‐3p in DD, miR‐221‐3p was overexpressed in CUMS rats and combined with silencing METTL3.…”

“…Dysfunctional miRNAs are associated with stress vulnerability, which further leads to an aggravation of DD symptoms (Allen & Dwivedi, 2020; Lopizzo, Zonca, Cattane, Pariante, & Cattaneo, 2019). Of note, miR‐221‐3p is found to be enriched in the serum and cerebrospinal fluid of DD patients and increased miR‐221‐3p indicates high depressive mood scores (Cui, Ma, Kong, & Song, 2021; Feng et al, 2019). In terms of its downstream mechanism, GRB2‐associated binding protein 1 (Gab1) was predicted to be a target of miR‐221‐3p by databases.…”

“…Most importantly, we established ROC curves for plasma microRNA-221-3p levels, and the results showed excellent performance for POCD identification. Previous studies have reported that microRNA-221-3p is related to the pathogenesis of many cerebrovascular diseases, the microRNA-221-3p levels in plasma and CSF can be used as risk and diagnostic indicators of PSD (poststroke depression), moreover, the upregulated plasma microRNA-133b and microRNA-221-3p levels may be biomarkers for early PD [ 26 ]. In addition, microRNA-221-3p plays a role in the autophagy of dopaminergic neurons in a mouse model of PD [ 27 ].…”

“…MMSE score was used to define cognitive recovery in one study at 14 days after stroke [58], whereas MoCA score assessed cognitive impairments in three studies in the chronic phase of stroke patients (within 1 year) [50,51,55]. To measure depression symptoms among post-stroke patients, three studies evaluated HAMD scores within 2-3 weeks and at 3 months after stroke onset [99][100][101].…”
