Diacylglycerols stimulate short-circuit current across frog skin by increasing apical Na+ permeability

Abstract: The phorbol ester TPA (12-O-tetradecanoylphorbol-13-acetate) stimulates baseline Na+ transport across frog skin epithelium and partially inhibits the natriferic response to vasopressin. The effects are produced largely or solely when TPA is added to the mucosal surface of the tissue. Although TPA activates protein kinase C, it has other effects, as well. Thus, the biochemical basis for the effects and the ionic events involved have been unclear. Furthermore, the physiologic implications have been obscure becau… Show more

“…A number of lines of indirect evidence have led us to suggest that protein kinase C is one mediator of insulin's stimulatory action of Na+ transport across frog skin (17). (i) Insulin (4) and activators of protein kinase C (17,19,22) stimulate Na+ transport across this tissue.…”

“…Indeed, although a large number of studies have examined ENaC's regulation by PKC in various preparations (3,7,11,15,24,27,30,32,33,39,42), only a minor fraction have focused on the potential regulation by distinct isoforms or a group of isoforms. Moreover, these studies utilized pharmacological agents believed to be selective for certain isoforms.…”

“…Insofar as Cd =+ is thought to reduce the apical C1-conductance of frog skin (Hayashi et al, 1977), these experiments were performed with skins bathed with the standard Ringer's solution on their serosal surfaces, but with an equimolar replacement of NO3 for Clon their mucosal surfaces. This approach has been empirically used in studies of frog skin to reduce the baseline, non-Na + conductance (Nagel, GarciaDiaz & Essig, 1983;Civan et al, 1987;Leibowich et al, 1988).…”
