Diabetic Nephropathy in KK and KK-Ay Mice.

Abstract: KK mice and KK-Ay mice were examined for age related changes in blood and urinary biophysiological parameters. Blood hemoglobin A1c levels were significantly higher in KK-Ay and KK mice as compared to non-diabetic ddY mice. In both diabetic mice, especially KK-Ay mice, plasma insulin levels markedly increased at 2 to 4 months of age, and the urinary glucose and microalbumin levels and albumin-to-creatinine ratios increased dependent on age. Plasma thrombomodulin levels significantly increased at 2 to 4 months … Show more

“…Albuminuria is a well known early sign of human DN. This clinical sign has already been confirmed in KK-A y mice [11,23], and the age-dependent gradual increase of the urinary albumin/creatinine ratio reported in these previous study also agreed well with the changes of glomerular sclerotic lesion scores seen in the present study.…”

“…The DN phenotypes in KK-A y mice are more severe than those in the original KK mouse [23], and a recent study showed that DN in KK-A y mice resembled human DN, both histopathologically and immunohistochemically [11]. An interaction between hemodynamic and metabolic factors induces DN [28].…”

“…Many mouse models of diabetes have been established including type 1 diabetes due to streptozotocin (STZ) treatment and genetic models (e.g., C57BL/ 6-Ins2 Akita /J) (12)(13)(14). Models of type 2 diabetes include diet-induced and inherited deficiency in the leptin receptor (i.e., db/db) mice (15)(16)(17). However, nephropathy has been studied in only a limited number of strains, and none of these models develops renal lesions that fully resemble advanced human diabetic nephropathy (18).…”

“…Okazaki et al (2002) reported that blood hemoglobin Al* Ievels were significantly correlated with urinary albumin excretion in KK-Ay mice. In this study, the mice excreted a significant amount of albumin in the urine; the amount was significantly smaller in the KK-Ay mice fed the 6-MSITC diet than in the controls.…”
