Developmental Validation of the SPERM HY-LITERTM Kit for the Identification of Human Spermatozoa in Forensic Samples*,†

Abstract: With sexual assault evidence, the visualization of spermatozoa confirms that ejaculation has occurred. However, microscopic examination of spermatozoa is a laborious process and can sometimes result in sperm cells being overlooked. Here, we present the developmental validation of the SPERM HY-LITER™ kit, which contains a human sperm-specific mouse monoclonal antibody coupled to a fluorescent Alexa 488 dye. The kit was tested using samples of human semen, saliva, blood, and urine, various animal semen extracts,… Show more

“…These results are consistent with Miller et al who confirmed that detection of human spermatozoa head is highly improved via using Sperm Hy-Liter  in comparison with other traditional staining methods, such as HE. This could be attributed to the antibody binding sites on the head of spermatozoa which are not deteriorated easily during the putrefaction [17]. The present study revealed that monoclonal antibody method outperforms SEM.…”

“…Liczba zidentyfikowanych plemników ma kluczowe znaczenie. Stanowi bowiem istotny wskaźnik, czy prób-ka zawiera wystarczającą ilość materiału do profilowania DNA w celu identyfikacji sprawcy przestępstwa [17]. Według jednego z doniesień pełne profile DNA uzyskano na podstawie analizy co najmniej 30 plemników [18].…”

“…Prostate-specific antigen, semenogelin, and MHS-5 in seminal fluid have also been detected by serological methods, including immunochromatography and ELISA [3][4][5][6][7][8][9][10]. However, these constituents are not specific to seminal fluid because they are also present in other body fluids such as urine, vaginal fluid, sweat, and body tissues [3,4,11,12]. Thus, the seminal markers described above can only be used as presumptive tests for semen.…”
