Developmental regulation of cytokeratins in cells of the rat mammary gland studied with monoclonal antibodies.

Abstract: We have isolated two monoclonal antibodies to cytokeratins and determined their cell specificities. They display interesting localization within the rat mammary gland. One (lA10) shows specificity for myoepithelial cells; the other (24B42) is specific for lumenal cells at various stages of development. These two monoclonal antibodies and three others to cytokeratin previously isolated were used in conjunction with antibodies to myosin and collagen IV to confirm and extend our previous findings on epithelial ce… Show more

“…Therefore, it also will be useful in defining the role and relationship of the various cytokeratins during myoepithelial and luminal epithelial development and differentiation. Precedence for such myoepithelium-specific keratins has been established in the mammary gland of rodents (12,13). However, the antibodies used in these studies do not crossreact with their human counterparts; and thus, they cannot be applied toward investigations of the human myoepithelium.…”

“…The presence of E cadherin in the domes probably indicates that it allows the formation of junctions of a special nature, enabling the cell layer to be lifted by the underlying hydrostatic pressure produced by the pumping function. The expression of a cytokeratin is also a sign of a differentiation process, because cytokeratins of various types are expressed in the cells of mammary ducts at various stages of differentiation (31). Cytokeratin 8, which is expressed in the domes, is frequently associated with tubular structures in other organs (32).…”
