The mass mortalities of sea cucumber Apostichpous japonicus have prevailed in northern China, mainly attributing to the emergence of extreme environmental conditions, that is hyperthermia and hyposalinity. The high-quality sea cucumber seedlings appear to possess more robust resistance to adverse conditions. There are usually indoor-and outdoor-cultured seedlings in industrial production of sea cucumbers.Although the outdoor-cultured sea cucumbers are practically considered to be more strong and robust, the effective evaluation approach to distinguish these seedlings has been scarce. The current study compared survival and immune performances of indoor-and outdoor-cultured A. japonicus under combined exogenous stressors, that is hyperthermia and hyposalinity. Results based on secondary stress induction revealed that the activities of immune enzymes and levels of catecholamines in body wall of outdoor-cultured seedlings were prominently higher than those of indoor-cultured seedlings recovered for 0-72 hr following sublethal stress (30°C and 25 psu of salinity). The opposite case occurred on immune enzymes in coelomic fluid of the two sources of seedlings except for myeloperoxidase. Importantly, the outdoor-cultured seedlings, which were recovered for 72 hr after sublethal stress, exhibited a 93% of cumulative survival rate following 7 days of recovery after lethal stress (33°C and 20 psu of salinity), 27% higher than the indoor-cultured seedlings. Collectively, the outdoor-cultured A. japonicus seedlings showed more superior quality than the hatchery-produced seedlings in terms of survival and immune performance. These findings provide practically useful information towards quality distinction of the indoor-and outdoor-cultured sea cucumbers, which could benefit the aquaculture industry to obtain high-quality seedlings. Aiming at the problem that the cleaning effect and surface integrity of the mechanical cleaning process cannot be solved simultaneously in the pretreatment of sea cucumber and according to the requirements of high efficiency, high quality and economy of sea cucumber body surface cleaning, a continuous directional cleaning device for sea cucumber abdomen and back based on the principle of water jet is designed. Through single factor experiment, the influence trend of jet parameters such as jet pressure, target distance and incidence angle on cleaning effect was analyzed. The cleaning parameters of sea cucumber are optimized by L9(3 4) orthogonal design. The experimental results show that the cleaning effect of sea cucumber is closely related to the impact of water jet. Enhancing the impact of water jet will not only help improve the cleanliness of sea cucumber, but also increase the risk of skin damage of sea cucumber. T he influence order of water jet parameters on the cleaning effect of sea cucumber is as follows: Jet pressure > jet target distance > jet incidence angle. When the jet pressure is 3.0 MPa, the jet target distance is 200 mm and the jet incidence angle is 70°, the cleanliness and integrity scores are 8.0 and 8.2 respectively and the comprehensive cleaning effect of this scheme is optimal. This study provides the basis for the research and development of sea cucumber water jet cleaning device and process optimization.

Landu beach is a beach located on Landu Island, Southwest Rote District, Rote Ndao Regency, which is the southernmost district in Indonesia. Sea cucumbers are used by the coastal community of Landu Beach as food and sold to the market as a source of income. So far, it is not scientifically known the types of sea cucumbers and the level of sea cucumber diversity in the coastal waters of Landu. Therefore, this study aims to identify the diversity and abundance of sea cucumbers on Landu Beach, Landu Island, Rote Ndao Regency. This study used a survey method with the sampling technique using the quadratic transect method (quadrant transect method). With 5 transects, each transect consists of 5 plots. The data taken were in the form of sea cucumber species, the number of sea cucumbers per plot, and the state of the substrate per observation plot. The results of the study found 4 types of sea cucumber species in Landu Beach, namely Holothuria scabra, Holothuria arata, Holothuria leucospilota, and Bohadchia marmorata. The abundance of each type of sea cucumber found was Holothuria Scabra with the highest abundance value, namely 2.44 ind / m2 followed by Holothuria Arta (2.16ind / m2), Bohadchia Marmorata (1.92 ind / m2), and Holothuria Leucospilota (1.68 ind. / m2). Bohadchia Marmorata species has a frequency of 1.0 which means this species was found in the research plot seedlings. The next frequency values are Holothuria Scabra (0.72), Holothuria Arta (0.68), and the lowest frequency value is Holothuria Leucospilota (0.60). For the diversity index value according to the Shannon-Wiener index (H ') ranges from 1.37 so it is categorized that Landu Beach has moderate diversity.
