Development of Sanitation Protocol for Leaf Explants of B. huillensis for in Vitro Culture

Abstract: Brachylaena huillensis (Silver Oak) is a multipurpose timber tree species in the family of Asteraceae. There has been a very high demand for B. huillensis wood and its products leading to overexploitation. B. huillensis regenerates through seedlings. However, it produces seeds with poor germination. Seeds are also difficult to be collected because of small size. Many are eaten by insects and currently there is a lack of seed bank. The facts that have hindered and rendered the natural regeneration of the tree s… Show more

“…The explants were immersed in 70% alcohol for 3 minutes and then rinsed using sterile distilled water three times. Afterwards, the leaf explants were immersed in 1.5% NaOCl for 15 minutes (Ndakidemi et al, 2013) and then rinsed using sterile distilled water three times. The sterilized explants were dried in sterilized filter paper on a petridish before further treatments.…”

“…In other words, the forest was rich with abundance of I. gabonensis species, but due to poverty, economic hardship, and human search for non-timber forest products as alternative means of survival, I. gabonensis species has likely been overexploited or affected by wildfire. Therefore, there is a need to carry out a natural regeneration status to ascertain its survival rate in the near future (Ndakidemi and Ndakidemi 2013). As such the study intends; to determine the abundance of I. gabonensis species with respect to age classes; seedlings, saplings, poles, and matured, to investigate the vegetative composition and also the status of natural regeneration on different ecological zones of the forest and to also determine indigenous knowledge on I. gabonensis by locals around the forest (Adekunle, 2007).…”
