Development of safe, effective and immunogenic vaccine candidate for diarrheagenic Escherichia coli main pathotypes in a mouse model

Abstract: BackgroundEnteric and diarrheal diseases are important causes of childhood death in the developing world. These diseases are responsible for more than 750 thousand deaths in children under 5 years old worldwide, ranking second cause of death, after lower respiratory diseases, in this age group. Among the major causative agents of diarrhea is Escherichia coli. There are several vaccine trials for diarrheagenic E. coli. However, diarrheagenic E. coli has seven pathotypes and vaccines are directed for one or two … Show more

“…Foodborne pathogens such as E. coli and other enterobacteria are essential causes of diarrhoeal diseases. Enteric and diarrheal diseases are important causes of childhood death in the low-and-middle-income countries; and are regarded as the second cause of death, after lower respiratory diseases in children under five years old [ 49 , 50 , 51 ]. The South African National Institute for Communicable Diseases (NCID) report of foodborne disease (FBD) for January to June 2017 indicated that Staphylococcus , Salmonella, and Clostridium perfringens were the leading cause of FBD in the period previously mentioned [ 52 ].…”

These diseases are responsible for more than 750 thousand deaths in children under 5 years old worldwide, ranking them as the second cause of death, after lower respiratory diseases, in this age group [ 2 ]. Diarrheal diseases are often caused by contaminated food: the first assessment of the global burden of foodborne disease estimated a burden of 600 million cases of foodborne illness and 420,000 deaths in 2010 [ 3 ].…”

“… 6 ). In one study, mice were immunized subcutaneously with five different pathogenic E. coli KWCVs ( 7 ) with no adverse effects noted. In another study, an oral genetically engineered enterotoxigenic E. coli (ETEC) vaccine overexpressing colonization factors was safe in mice ( 8 ).…”
