Development of recombinant stable house dust mite allergen Der p 3 molecules for component‐resolved diagnosis and specific immunotherapy

Abstract: rDer p 3-S196A should be used for the diagnosis of HDM allergy elicited by Der p 3, and proDer p 3 may represent a hypoallergen of Der p 3.

“…12 Recently similar low binding was reported for natural and functional rDer p 3 but a noncatalytic mutant showed higher binding for some sera providing a valuable reagent for further study. 13 The Der f 15 14 &18 15 allergens were discovered as important allergens of HDM-allergic dogs having primary structures similar to chitinase enzymes. It is however now known that the highly reactive and heavily glycosylated Der f 15 bound dog IgE via carbohydrate determinants 16 and that the IgE binding by humans found in 40% of allergic subjects is usually of low titre.…”

“…The natural group 7 allergens can be highly glycosylated (30%) and there are at least 3 different glycosylation isoforms found in varying degrees in different HDM extracts. 50 Minor allergens (groups 3,6,8,9,10,11,13,15,16,17,18,20) (Table 3) Group 3, 6 &9 allergens…”

“…Not all whole allergen preparations contain adequate amounts of all allergen components, and Der p 3 is one such component. Bouaziz et al [94] described a synthetic form of this allergen for use to improve the diagnosis of house dust mite allergy. Other cross-reactive allergens can also impede the diagnostic value of a test, and one example was described in a research letter from Walton et al [95].…”

“…Der p 1 has also been demonstrated to act as the activator of the precursors of Der p 3 and Der p 6 according to an uncommon activation cascade [ 5 , 6 , 7 ]. In contrast to Der p 1, little is known about the concentration of Der p 3 and Der p 6 in mite cultures and their IgE binding frequencies remain poorly characterized with IgE prevalence ranging from 10% to 50% for Der p 3 and around 40% for Der p 6 [ 8 , 9 , 10 ].…”
