Development of new generation of vaccines for Brucella abortus

Abstract: Brucella abortus is a Gram-negative facultative and intracellular bacteria, it causes bovine brucellosis, a zoonotic disease that is responsible for considerable economic loss to owners of domesticated animals and can cause problems in otherwise healthy humans. There are a few available live attenuated vaccines for animal immunization against brucellosis; however, these have significant side effects and offer insufficient protective efficacy. Thus, the need for more research into the Molecular pathobiology and… Show more

“…There are genes associated with virulence or organism survival; research into these genes will aid in the development of safe and protective vaccines, such as live but attenuated vaccines, which provide significant amounts of protection [ 70 ]. Purine biosynthesis pathway genes ferro chelatase hem H mutant, a gene required to transport lipid A 363 fatty acid, the phosphoglycerate kinase-encoding gene, LPS biosynthesis pathway genes, and 364 Type IV secretion Vir B genes are just a few examples of vaccines developed using deletions in B. abortus virulence genes that result in major attenuation [ 71 , 72 , 73 , 74 , 75 , 76 ].…”

“…Although S19∆per has the added advantage of the safety parameters, to be considered as a potential vaccine candidate, it needs to commensurate with the inherent ability of conferring protection against infection. The LPS deficient mutants have been demonstrated to be highly attenuated in a number of studies, but in many instances, they failed to induce protective immune responses [47,48].…”

“…B. abortus strains 19 and RB51 are very efficient and common vaccines being used against bovine brucellosis. B. abortus S19 was a result of natural attenuation lacking 720-bp region in the erythritol catabolic genes (Sangari et al 1994 ; reviewed in Gheibi et al 2018 ). The strain RB51 vaccine does not interfere with serodiagnostic results unlike strain 19 vaccine (Moriyón et al 2004 ).…”
