Mushrooms have been accepted as nutraceutical foods because of their high nutritional and functional values. They have also gained interest due to their medicinal properties, economic importance, and organoleptic merit. In this study, wild Ganoderma lucidum and four commercial mushrooms, that is, Pleurotus ostreatus, Volvariella volvacea, Hericium erinaceus, and Lentinus edodes from Pakistan were screened for their biological activities such as anticancer, antityrosinase, anti‐α‐glucosidase, and antithrombotic activities from their methanol, ethanol, and water extracts. Enzyme inhibition assay showed that selected mushrooms are potent inhibitors with %age inhibition ranging from 19.00% to 80.91%, and 32.85% to 83.38% for tyrosinase and α‐glucosidase, respectively. The best tyrosinase inhibition was shown by P. ostreatus whereas L. edodes was found best as α‐glucosidase inhibitor. These mushrooms were tested against cancer cell lines including HT‐29 colon and H‐1299 lungs carcinoma cell lines. G. lucidum showed 29% and 24% viability of cells against HT‐29 and H‐1299 cell lines, respectively. This antiproliferative effect was in dose‐dependent manner, and the maximum inhibition was observed at 200 μg/ml. Mushrooms extracts were also found effective against clot lysis. The percentage of clot lysis was in the range of 27%–29%. The research would provide knowledge to the people of Pakistan about the importance of locally available commercial mushrooms and wild mushrooms for health improvement and prevention against different kinds of diseases.

Wilson (Verbenaceae) and Zanthoxylum rhoifolium Lam (Rutaceae) ABSTRACT Cecropia hololeuca, Lippia alba, and Zanthoxylum rhoifolium are plant species naturally occurring in Brazil, from Atlantic Forest, with medicinal purposes, like antihypertensive action, digestive, and treatment of malaria, respectively. In this study, we evaluated the chemical profile, cytotoxicity, and thrombolytic potential of the ethanol extract of the leaves of these plant species. The phytochemical profile was characterized by phytochemical tests of metabolites classes identification. The preliminary toxicity of the extracts was determined by brine shrimp lethality bioassay and the in vitro thrombolytic activity was evaluated through the extracts capacity to cause lysis in human blood clot. Phytochemicals tests indicated the presence of flavonoids and alkaloids in the three extracts. Saponins, triterpenes, and naphthoquinones were only positive for C. hololeuca. Cecropia hololeuca did not show cytotoxicity to Artemia salina (LD 50 >1000 ppm) while Z. rhoifolium presented LD 50 equal to 719.44 ppm and L. alba <250 ppm, which indicates attention to the safety of this medicinal plant use. The thrombolytic activity of L. alba and C. hololeuca was 6.43 ±2.08 and 9.64 ±1.83 %, respectively, showing a small potential. Z. rhoifolium lysis achieved 24.71 ±10.52%, indicating promising activity (p <0.001). The thrombolytic activity of Z. rhoifolium justifies further studies to investigate the components responsible for this activity. This study, faced with the literature, is the first report assessing the thrombolytic activity of these plant species. The findings of this study contribute to the chemical-biological knowledge of these species. The comparative study was aimed to evaluate the potential anticancerous and cardioprotective activities of methanolic leaf extract of Cycleabarbata and Entadapursaetha. The screening of anticancerous activity was performed using brine shrimp lethality bioassay while the cardioprotective activity was evaluated using the in vitro clot lysis model. Significant anticancerous activity was found for methanolic leaf extract of C. barbata whereas moderate effect was found for methanolic leaf extract of E. pursaetha and it was compared with the standard drug vincristine sulfate in the brine shrimp lethality bioassay. In the present study, the LC 50 values of methanolic crude extract of C. barbata, E. pursaetha and vincristine sulfate were 104.04, 330.31 and 12.59 μg mLG 1 , respectively. In cardioprotective study, it was found that C. barbata and E. pursaetha showed 45.69±1.96 and 38.19±1.83% of clot lysis, respectively. Between the plants studied C. barbata, showed very significant (p<0.05) percentage of clot lysis than E. pursaetha compared to reference drug streptokinase which is 63.54±2.61%. The results of this study demonstrated that the leaf of these plants possess promising anticancerous activity on brine shrimp as well as significant cardioprotective activity in vitro when tested on human blood.
