. Significance : Diffuse optical tomography is an ill-posed problem. Combination with ultrasound can improve the results of diffuse optical tomography applied to the diagnosis of breast cancer and allow for classification of lesions. Aim : To provide a simulation pipeline for the assessment of reconstruction and classification methods for diffuse optical tomography with concurrent ultrasound information. Approach : A set of breast digital phantoms with benign and malignant lesions was simulated building on the software VICTRE. Acoustic and optical properties were assigned to the phantoms for the generation of B-mode images and optical data. A reconstruction algorithm based on a two-region nonlinear fitting and incorporating the ultrasound information was tested. Machine learning classification methods were applied to the reconstructed values to discriminate lesions into benign and malignant after reconstruction. Results : The approach allowed us to generate realistic US and optical data and to test a two-region reconstruction method for a large number of realistic simulations. When information is extracted from ultrasound images, at least 75% of lesions are correctly classified. With ideal two-region separation, the accuracy is higher than 80%. Conclusions : A pipeline for the generation of realistic ultrasound and diffuse optics data was implemented. Machine learning methods applied to a optical reconstruction with a nonlinear optical model and morphological information permit to discriminate malignant lesions from benign ones.

We present two optical breast atlases for optical mammography, aiming to advance the image reconstruction research by providing a common platform to test advanced image reconstruction algorithms. Each atlas consists of five individual breast models. The first atlas provides breast vasculature surface models, which are derived from human breast dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) data using image segmentation. A finite element-based method is used to deform the breast vasculature models from their natural shapes to generate the second atlas, compressed breast models. Breast compression is typically done in X-ray mammography but also necessary for some optical mammography systems. Technical validation is presented to demonstrate how the atlases can be used to study the image reconstruction algorithms. Optical measurements are generated numerically with compressed breast models and a predefined configuration of light sources and photodetectors. The simulated data is fed into three standard image reconstruction algorithms to reconstruct optical images of the vasculature, which can then be compared with the ground truth to evaluate their performance.

This research was intended to explore the application value of photoacoustic imaging technology based on image intelligent iterative reconstruction algorithm in the detection and diagnosis of early oral cancer. An iterative algorithm model was constructed and systematically analyzed. The algorithm was used to debug the detection of B-scan images on the diameter of the imaging area. The results showed that the sensitivity of line-focused ultrasound detector was 86.72% and the specificity was 80.79%, while the sensitivity of the flat-field ultrasound detector was 63.15% and the specificity was 71.79%. The photoacoustic microscopy imaging technology can clearly observe the rich capillary network on human lips. A part of the vascular network at the depth of 100 μm, 500 μm, and 1000 μm grew out of the reticular capillaries and extended out of the loop-like capillaries, and the diameter gradually expanded. The imaging experiment of the sublingual capillary network in the human body showed that loop-like capillaries were observed, but there were some large blood vessels, which corresponded to the densely distributed blood vessel network under the tongue. The morphological changes of loop-like capillaries can be well observed by photoacoustic microscopy. In conclusion, the reconstructed photoacoustic microscopy imaging technology can realize high-resolution imaging of human oral capillaries and observe the morphological changes of loop-like capillaries, which had a certain application value for the early detection of oral cancer.
