Development of a Class-Specific ELISA for Sulfonylurea Herbicides (Sulfuron Screen)

Abstract: The development of a direct competitive ELISA for the detection of a broad range of sulfonylurea herbicides (SUs) is described. Polyclonal antibodies were generated in rabbits using three different immunizing haptens. Antiserum with the broadest specificity was obtained with a mesosulfuronbenzylamine derivative which was coupled via a succinic acid spacer to keyhole limpet hemocyanine. A heterologous enzyme tracer which did not contain the succinic acid bridge was prepared using activated horseradish peroxidas… Show more

“…The antigen-antibody biochemical interaction could often be affected by a variety of compounds, such as inorganic salts and humic substances, etc. 28,29 In a previous study where an ELISA was used for the detection of butachlor, without dilution treatment, matrix effects were found in tap water, pond water, river water, bottled water and rice field water, and no obvious matrix effect was only found for distilled water. 30 However, in the present FPIA, recoveries were satisfactory for several water samples except for a small matrix effect in pond water.…”

“…It is a big challenge for us to enrich and separate TBM, for its extremely low residual as well as being mixed with complex matrix. Up to now, different methods have been developed to determine TBM residues, including high performance liquid chromatography with ultraviolet detector (HPLC-UV) [5], high performance liquid chromatography with mass spectrum (HPLC/MS/MS) [6, 7], capillary electrophoresis (CE) [810], and enzyme-linked immunoassay (ELISA) [11]. Along with these methods, most sample pretreatments for sulfonylurea analysis used solid-phase extraction (SPE) with C 18 columns for purification and concentration.…”

“…A variety of instrumental methods have been proposed for the quantitative analysis of SUs, including ELISA , CE , and HPLC . HPLC with UV detection or diode array detection (DAD) has been the most frequently used technique for the analysis of these polar and thermally labile herbicides.…”
