In this study, the use of weighted linear regression in the development of electrochemical methods for the determination of epinephrine (EP), ascorbic acid (AA), and uric acid (UA) is presented. The measurements were performed using a glassy carbon electrode and square-wave voltammetry (SWV). All electroanalytical methods were validated by determination of the limit of detection, limit of quantification, linear concentration range, accuracy, and precision. The normal distribution of all data sets was checked using the quantile-quantile plot and Kolmogorov-Smirnov statistical tests. The heteroscedasticity of the data was tested using Hartley’s test, Bartlett’s test, Cochran’s C test, and the analysis of residuals. The heteroscedastic behavior was observed with all analytes, justifying the use of weighted linear regression. Six different weighting factors were tested, and the best weighted model was determined using relative percentage error. Such statistical approach improved the regression models by giving greater weight on the values with the smallest error and vice versa. Consequently, accuracy of the analytical results (especially in the lower concentration range) was improved. All methods were successfully used for the determination of these analytes in real samples: EP in an epinephrine auto-injector, AA in a dietary supplement, and UA in human urine. The accuracy and precision of real sample analysis using best weighted model gave satisfactory results with recoveries between 95.21–113.23% and relative standard deviations between 0.85–7.98%. The SWV measurement takes about 40 s, which makes the presented methods for the determination of EP, AA, and UA a promising alternative to chromatographic techniques in terms of speed, analysis, and equipment costs, as the analysis is performed without organic solvents.

Thunbergia laurifolia has been a popular herb used in Thai traditional medicine for detoxification and as antipyretic. It contains rosmarinic acid (RA), caffeic acid (CA) and vitexin as major compounds. In order to control the herbal quality, the stability indicating high-performance liquid chromatography (HPLC) was developed and validated. The stability study of compounds in T. laurifolia leaf extract was investigated. The chromatographic separation was performed using a reversed-phase C18 column and mobile phase consisted of 0.5% acetic acid and methanol using a gradient elution with 1.0 mL/min flow rate. The detection wavelength was set at 330 nm. The method was validated for its linearity, precision, accuracy, limit of detection and limit of quantitation. Forced degradation of three compounds in extract showed that they were stable in oxidative condition, but highly labile under alkaline hydrolytic conditions. All three compounds in T. laurifolia leaf extract were stable at room temperature at least 3 months while a remarkable decrease of RA, vitexin and CA in the extract were found in accelerated condition. This finding could be applied for predicting the storage recommendation and expiry of T. laurifolia extract and its related pharmaceutical products.

A reliable RP-HPLC analytical method with UV detection at 421 nm was developed and validated for the quantitative determination of curcumin from rat plasma after oral administration of curcumin loaded nanocochleates (CU-NC) to rats. The chromatographic separation was performed on HIQ SIL, C18 (250 mm × 4.6 mm) column using methanol and water (80:20 v/v) as mobile phase, at 1.0 mL/min flow rate. Validation parameters included linearity, accuracy, precision, and limit of quantitation and detection. Good linearity was obtained over the range of 2.5–100 μg/mL (R2 = 0.9979) of curcumin. The developed HPLC method was precise, with <2% relative standard deviation. Accuracy, stability, and robustness studies were also found to be acceptable. Bland-Altman plot showed an acceptable repeatability coefficient. The method was under statistical control, revealed by a control chart. After CU–NC administration, pharmacokinetic parameters i.e. Cmax, AUC0-∞, and AUMC0-∞, were observed to be 97.69 ± 10.84 μg/mL, 1402.77 ± 9.67 (μg/mL) ∙ h, and 35140.16 ± 14.67 (μg/mL) ∙ h2, respectively. This simple and precise method can be effectively implemented for routine analysis.
