Development and Validation of RP-HPLC Method for Simultaneous Estimation of Glimepiride and Sildenafil Citrate in Rat Plasma-Application to Pharmacokinetic Studies

Abstract: A simple and sensitive method was developed for simultaneous estimation of Glimepiride (GLIM) and Sildenafil citrate (SIL) in rat Plasma by reverse phase high performance liquid chromatography (RP-HPLC). The drug samples were extracted by liquid-liquid extraction with 300 µl of acetonitrile and 5 ml of diethyl ether. Chromatographic separation was achieved on C18 column using methanol: water (85:15 v/v) as mobile phase at a flow rate of 1 ml/min and UV detection at 230 nm. The retention time of GLIM and SIL wa… Show more

“…Many quantitative methods have been developed for the determination of GLM in biological fluids, such as high-performance liquid chromatography with ultraviolet detection (HPLC-UV) which showed, relatively, very high quantitative limits, that are far from the actual amount of non-metabolized GLM [6][7][8][9]. Also, liquid chromatography-tandem mass spectrometry (LC-MS) has been developed for its determination [10][11][12][13].…”

“…Also, liquid chromatography-tandem mass spectrometry (LC-MS) has been developed for its determination [10][11][12][13]. Generally, two extraction procedures were used for the recovery of GLM from plasma, including liquid-liquid extraction (LLE) [9] and solid-phase extraction (SPE) [8,10]. The major limitation of LLE that it consumes a large quantity of organic solvent and leads to environmental pollution.…”
