Development and validation of an enzyme-linked immunosorbent assay for detection of cortisol in human saliva

Abstract: Non-invasive measurement of cortisol in saliva is of prime importance as it represents a bioavailable neuroendocrine marker for stress. Therefore, in this study, we developed an enzyme immune assay that was suitable for salivary cortisol measurements. For that purpose, rabbit polyclonal antibody was raised against cortisol-3-CMO:BSA conjugate. The test was based on competition of liquid phase cortisol with conjugated cortisol on the solid phase. Primary antibody was used to bind available sites on the conjugat… Show more

“…Cortisol level, which is one of the biological markers of stress, is determined by means of the salivary cortisol level. Saliva samples were collected according to the expert consensus guidelines reported by Stalder et al [ 19 ] and following the rules explained by Ozgocer et al [ 20 ] Experimental procedure and study protocol were briefly explained to the participants during face-to-face interview. Polypropylene tubes (1.5 mL, ISOLAB Laborgeräte GmbH, Wertheim, Germany) labeled with the participant’s ID and sample number (e.g.…”

“…Following thawing, samples were centrifuged at 4000 g for 10 min and the supernatant was used for ELISA analyses as reported by Ozgocer et al. (2017b). Samples were diluted 5x with assay buffer and assayed in triplicate.…”

“…After incubation, stop solution (50 μl/well) was added and the absorbance was measured at 450 nm using a microplate reader. İntra‐assay variation (CV) is % 7.8 and inter‐assay variation is % 7.2 (Ozgocer et al, 2017b).…”

“…As a strict routine before the surgery, the children were not permitted to eat or drink before the surgery. Salivary cortisol levels were analyzed by a validated ELISA (enzyme-linked immunosorbent assay) method reported by Ozgocer et al (10). Of 87 cases, 41 did not receive premedication and 46 received hydroxyzine.…”
