Development and validation of a GC–FID method for quantitative analysis of oleic acid and related fatty acids

Abstract: Oleic acid is a common pharmaceutical excipient that has been widely used in various dosage forms. Gas chromatography (GC) has often been used as the quantitation method for fatty acids normally requiring a derivatization step. The aim of this study was to develop a simple, robust, and derivatization-free GC method that is suitable for routine analysis of all the major components in oleic acid USP-NF (United States Pharmacopeia-National Formulary) material. A gas chromatography–flame ionization detection (GC–F… Show more

“…The percent recovery of each ALA isomer standard added to heated trilinolenin is shown in Table . High recovery percentages of 96.2–106.5%, with CVs ranging from 0.82% to 0.97%, were attained for all ALA isomer standards and were much higher than those reported in a previous study by Zhang et al , who analyzed the 9 Z ,12 Z ,15 Z standard content and reported the recovery to be approximately 93%. Recovery values showed that the derivatization method used to determine ALA isomers in samples was appropriate.…”

“…In this study, results obtained by both methods (B&D and SOXTEC) are comparable with data from the aforementioned studies. Among the most widely used methods for fat extraction from fish, there is B&D (18,19) and SOXTEC is another method used to extract fat not only from fish but from different matrixes; both methods are considered accurate and precise (13). When comparing the fat extraction from fish methods, the CRM recovery percentage was taken as indicator.…”
