Development and Scale-Up of Three Consecutive Continuous Reactions for Production of 6-Hydroxybuspirone

Abstract: This paper describes the development of a continuous, high yielding, and scalable enolization, oxidation, and quench process for the hydroxylation of the azapirone psychtropic agent buspirone to afford 6-hydroxybuspirone (6-hydroxy-8-[4-(4-pyrimidin-2-yl-piperazin-1-yl)-butyl]-8-aza-spiro[4.5]decane-7,9-dione). Two feed streams were reacted continuously using an in-line static mixer followed by oxidation in a continuous flow trickle-bed reactor. The laboratory reactor operation was demonstrated at steady state… Show more

“…Chemists from Bristol-Myers Squibb applied continuous flow technology to develop a scalable, high-yielding route for the hydroxylation of buspirone (7) by an enolization/oxidation sequence (Scheme 5) [45]. In the first step, the substrate feed was mixed with the base by slowly reducing the temperature in two sequential static mixing units (SMU) to avoid precipitation of the inorganic base.…”

“…A numbering-up technique is used for the oxidation step (quad reactor). Adapted with permission from [45] Aerobic Oxidations in Continuous Flow control of its performance. Notably, a constant purity profile was monitored (90%) over time at a high production rate (15 kg d À1 ).…”

“…Academic laboratories have been employing this technology as routine work [33,34] , in industry there is clearly considerable interest in the introduction of continuous processing [34] , for example, chemical companies such as Bayer [10] , AstraZeneca [4] , Bristol-Myers Squibb [35] , Lonza [11] and others [36] .…”
