RESUMOO protetor solar é um cosmético que tem como objetivo principal proteger a pele dos efeitos nocivos da radiação solar. A radiação UVA (ultravioleta A), por penetrar mais profundamente a pele é geralmente a maior responsável pelo fotoenvelhecimento e carcinogênese, enquanto que a radiação UVB (ultravioleta B) é considerada mais eritematosa. Dentre muitos ingredientes naturais estudados para promover o efeito fotoprotetor, o açafrão aparece como um ativo promissor, pois reúne diversas propriedades (filtro químico, antitumoral, antioxidante, anti-inflamatório). Nesse contexto, o presente trabalho teve como objetivo avaliar a ação fotoprotetora do componente majoritário dos rizomas do açafrão, a curcumina. Para tanto foram desenvolvidas loções cremosas fotoprotetoras contendo curcumina (1 e 2%), tanto na forma isolada quanto associada aos filtros químicos octilmetoxicinamato (UVB) e benzofenona-3 (UVA). A partir destas formulações foram feitas análises por espectrofotometria de varredura na região do ultravioleta (290 -400 nm) e determinação do FPS (fator de proteção solar) in vitro pelo método espectrofotométrico de Mansur et al. (1986). Os resultados mostraram que a curcumina apresenta absorção na região do UVAlongo, mais especificamente de 350 -400 nm, que não é contemplada pela mistura de filtros químicos utilizada. Em relação à determinação de FPS, que mede a proteção UVB fornecida pela formulação, o uso da curcumina de forma isolada não demonstrou ser eficiente (FPS = 0,70). A formulação contendo apenas curcumina apresentou um FPS incompatível com a possibilidade de sua comercialização como fotoprotetor (FPS = 6) ou produto multifuncional (FPS = 2). Quando associada aos demais filtros (FPS = 16,46; FPS = 16,21), não houve significativa variação do FPS em relação à formulação contendo apenas filtros químicos (FPS = 16,75). Deste modo, é possível concluir que a associação da curcumina com os filtros químicos sintéticos apesar de não aumentar muito o FPS, é benéfica, pois, resulta em uma formulação que garante uma proteção de amplo espectro, abrangendo inclusive a faixa do UVA-longo, região de difícil alcance por parte da maioria dos filtros químicos convencionais.Palavras-chave: fotoprotetor, açafrão, curcumina, FPS.

The present work evaluated the encapsulation of ZnO and quercetin in PBMA-PMMA-PS using the miniemulsion polymerization technique, aiming to develop nanoparticles with sun protection factor (SPF) and antioxidant activity (AA) for application in photoprotective lotions. In both formulations, octocrylene and green coffee oil were tested as co-stabilizing agents of the miniemulsion, being also encapsulated in the NPs, contributing to the SPF and AA of the obtained latexes. Spherical nanoparticles of homogeneous size, from 169 to 346 nm, and regular surfaces were obtained, remaining stable for at least 30 days. The encapsulation efficiency on the formulations tested was from 59 to 87% for ZnO, 47 to 51% for quercetin, 80 to 92% for octocrylene and 90 to 92% for green coffee oil. The quercetin and green coffee oil presented high antioxidant activity when encapsulated in polymeric NPs. The values of in vitro SPF was very good for formulations containing NPs-ZnO, with the best result for the simultaneous nanoencapsulation of ZnO and octocrylene (SPF 29±5). The application of the NPs of quercetin and green coffee oil may promote an increment on SPF in vivo, reducing the damage caused to the skin by UV radiation, beyond the ability to scavenge the free radicals generated by ZnO.

Curcuma longa L. is a rhizome plant often used as traditional medicinal preparations in Southeast Asia. The dried powder is commonly known as cure-all herbal medicine with a wider spectrum of pharmaceutical activities. In spite of the widely reported therapeutic applications of C. longa, research on its safety and teratogenic effects on zebrafish embryos and larvae is still limited. Hence, this research was aimed to assess the toxicity of C. longa extract on zebrafish. Using a reflux flask, methanol extract of C. longa was extracted and the identification and quantification of total flavonoids were carried out with HPLC. Twelve fertilized embryos were selected to test the embryotoxicity and teratogenicity at different concentration points. The embryos were exposed to the extract in the E3M medium while the control was only exposed to E3M and different developmental endpoints were recorded with the therapeutic index calculated using the ratio of LC50/EC50. C. longa extract was detected to be highly rich in flavonoids with catechin, epicatechin and naringenin as the 3 most abundant with concentrations of 3,531.34, 688.70 and 523.83μg/mL respectively. The toxicity effects were discovered to be dose-dependent at dosage above 62.50μg/mL, while at 125.0μg/mL, mortality of embryos was observed and physical body deformities of larvae was recorded among the hatched embryos at higher concentrations. Teratogenic effect of the extract was severe at higher concentrations producing physical body deformities such as kink tail, bend trunk, enlarged yolk sac edema. Finally, the Therapeutic Index (TI) values calculated were approximately same for different concentration points tested. Overall, the result revealed that plants having therapeutic potential could also pose threats when consumed at higher doses especially on the embryos. Therefore, detailed toxicity analysis should be carried out on medicinal plants to ascertain their safety on the embryos and its development.
