Development and Characterization of New Microsatellite Markers for the Oyster Mushroom (Pleurotus ostreatus)

Abstract: We developed and characterized 36 polymorphic microsatellite markers for the oyster mushroom (Pleurotus ostreatus). In total, 169 alleles were identified with an average of 4.7 alleles per locus. Values for observed (H O) and expected (H E) heterozygosities ranged from 0.027 to 0.946 and from 0.027 to 0.810, respectively. Nineteen loci deviated from Hardy-Weinberg equilibrium. Significant (P<0.05) excess heterozygosity was observed at nine loci. Linkage disequilibrium (LD) was significant (P<0.05) between pair… Show more

“…Previous studies of the genetic diversity of other macromycetes using genomic-SSRs reported an average of 4.7e5.6 alleles per locus with average genetic diversity values of 0.35e0.4. In this study, we observed a similar average of 9.875 alleles per locus and a higher average gene diversity value of 0.544 using genic SSR markers (Foulongne-Oriol et al, 2009;Ma et al, 2009). This may be due to the deep sequencing data we obtained.…”

“…Several reports on SSR used in Suillus bovines, Pleurotus ostreatus, and Agaricus bisporus, confirmed that microsatellite markers were essentially useful for genetic studies on mushroom (Kikuchi et al 2007;Foulongne-Oriol et al 2009;Kyung-Ho et al 2009). So, it is necessary to set up such an excellent marker technique in L. edodes, an economically important mushroom in the world.…”
