“…The protein was purified under denaturing conditions with the HIS‐select™ nickel affinity gel (Sigma, St. Louis, MO, USA, https://www.sigmaaldrich.com) according to protocol and dialysed against 4 M urea. After dialysis, Bac‐rCul o 3 was analysed with SDS‐PAGE and the gel stained with Coomassie blue, (PhastGel Blue R, Sigma) using standard protocols (Figure ) or transferred to a PVDF membrane (Millipore, Hellerup, Denmark, http://www.merckmillipore.com) and detected with mouse anti‐His antibody (Bio‐Rad, Hercules, CA, USA, https://www.bio-rad-antibodies.com), diluted 1:1,000, followed by the standard procedure of Western blot, as described previously …”

“…Allergen‐specific serum IgE levels against the ten recombinant C. nubeculosus allergens and NubTE, as well as against the three C. obsoletus r‐allergens and ObsWBE were determined by indirect ELISA. Levels of serum IgG subclasses against three of the most relevant rCulicoides allergen, rCul n 4, rCul o 1, rCul o 3, and NubTE and equine Influenza, were determined by ELISA, as described previously . Briefly, microtitre plates were coated with 2 μg/mL allergens, followed by blocking with blocking buffer (Phosphate buffered saline containing 5% dry milk powder and 5% Tween‐20).…”

“…IgG1 and IgG4/7 are thought to be essential for protection against viral and bacterial infections while the IgG3 and IgG5 have been associated with Th2 response and extracellular pathogens [22]. A high IgG4/7 background to E. coli expressed proteins has been observed previously in sera from some horses [14] and may be due to IgG4/7 antibodies against trace amounts of contaminating E. coli proteins present in the recombinant protein (purity of 95% [4]). This was also the case for IgG4/7, probably due to the fact that the pre-treatment sera showed very high IgG4/7 background on the E. coli-rCul n 2, used in the ELISA.…”

“…It is a reaction towards allergens present in the saliva of these biting midges. IBH has been described as a type I hypersensitivity reaction with release of histamine and other inflammatory mediators from basophils and mast cells (Jonsdottir et al, 2015). However the involvement of a type IV hypersensitivity reaction has also been suggested (Jonsdottir et al, 2015;Schaffartzik et al, 2012).…”
