Determination of Thiocyanate in Plasma and Saliva without Deproteinisation and Its Validation as a Smoking Parameter

Abstract: Summary: A simple photometric procedure was developed for the determination of thiocyanate (SCN~) in plasma and saliva without deproteinisation or dialysis. Fe(III) ions form a red coloured complex with SCNw ith a inaximum absorbance at 460 nm. Mercury(II) nitrate is used to run a sample blank. A manual and an automated Version (COBAS ΒΙΟ) of the method is described. The method is linear up to 5000 μηιοΐ/ΐ SCN~. The CV of the between^run precision is 2.8 -8% for the manual and 2.6 -6.6% for the automated metho… Show more

“…These findings were similar to a study of male Chinese smokers (Chu et al, 2009). Tobacco smoke contains high concentrations of hydrogen cyanide gas, which is primarily metabolized to thiocyanate, and serum thiocyanate was potentially a better biochemical index of smoking status (Degiampietro et al, 1987). The L allele of 5-HTTLPR likely expresses a higher number of 5-HTT sites on the serotonergic synapses (Heils et al, 1997), and this could result in higher serotonin reuptake activity, which could lead to decreased serotonin availability in the synaptic cleft.…”

“…Glucose, total cholesterol, triglycerides, high-density lipoprotein cholesterol (HDL-C), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were measured using enzymatic methods by DADE Dimension ® AR. Serum thiocyanate concentrations were assessed using the colorimetric method (Degiampietro et al, 1987). Low-density lipoprotein cholesterol (LDL-C) was estimated by the Friedewald formula, as follows:…”

“…were substantiated by measurement of thiocyanate in maternal and cord serum by a manual method (33 (14). Iodine in urine and breast milk was measured by the colorimetric method of the Sandell-Kolthoff reaction after alkaline ashing, as described previously (34).…”

“…WBC, RBC, and platelet counts and hemoglobin and hematocrit concentrations were also determined (Coulter Counter). Serum thiocyanate concentrations were assessed using the colorimetric method (Degiampietro et al, 1987). Serum BDNF levels were determined using an enzyme-linked immunosorbent assay protocol according to the manufacturer instructions (DBD00; R & D Systems).…”
