Determination of the triterpene glycosides in sea cucumbers by liquid chromatography with evaporative light scattering and mass spectrometry detection

Abstract: Holothurian triterpene glycosides possess various kinds of biological activities, including antifungal, cytotoxic, hemolytic, cytostatic, and immunomodulatory effects. In this study, a rapid extraction method of triterpene glycosides from sea cucumbers using a small column of C18 solid phase was first developed. Furthermore, a novel high-performance liquid chromatography method coupled with evaporative light scattering detection and electrospray ionization mass spectrometry was established for the determinatio… Show more

“…CAD is a universal detector suitable for weak or non‐UV absorption compounds, which have been developed to overcome the limitations of traditional HPLC detection. HPLC–ELSD was used for the characterization and quantitation of triterpene glycosides in sea cucumbers , triterpenoid saponins in raw and sprouted Chenopodium berlandieri spp. , rosmarinic acid, maslinic acid, corosolic acid, betulinic acid, OA, and ursolic acid in the extracts of Prunella vulgaris L., due to its capacity for the sensitive detection of nonvolatile compounds containing weak chromophores .…”

“…Qualitatively of triterpene glycoside was analyzed by high-performance liquid chromatography (also used to triterpenoid assay with added 50-100 mg/ml of triterpene glycoside in 0.5 ml of glacial acetate acid and incubated 15 min in tubes, and then added strong sulfate acid 0.5 ml). Positive has triterpene glycoside emerged brownish-red and purple colors [20,21].…”
