Determination of the components of analgesic mixtures using high-performance thin-layer chromatography

Abstract: A spectrodensitometric method is described for the determination of the components of two analgesic mixtures. For the first mixture (paracetamol-ascorbic acid-caffeine-phenylephrine), the pharmaceutically active components were separated from each other and closely related degradation products and impurities on high-performance thin-layer chromatography (F254) plates using methylene chloride-ethyl acetate-ethanol-formic acid (3.5 + 2 + 4 + 0.5) and methylene chloride-ethyl acetate-ethanol (5 + 5 + 1) as the de… Show more

“…Numerous methods have been used for the determination of paracetamol in pharmaceutical formulations and biological fluids including titrimetry [5,6], UV-Vis spectrophotometry [7][8][9][10], spectrofluorimetry [11], near infrared transmittance spectroscopy [12], electrochemical methods [13,14], and chromatography [10,[15][16][17][18][19]. Recently, several flow injection (FI) methods for the determination of paracetamol have been proposed using an UV-Vis spectrophotometer [20][21][22], fluorimeter [23], multioptosensor [24], or Fourier transform infrared spectrophotometer [25] as a detector.…”

“…El Sadek et al (44) describe an HPTLC method for the determination of the components of an analgesic mixture and used silica gel plates with methylene chloride-ethyl acetate-ethanolformic acid (3.5:2:4:0.5) as mobile phase for separating paracetamon, ascorbic acid, caffeine, and phenylephrine. The plates were scanned with a scanning spectrophotometer at 264 nm for ascorbic CH Ascorbic acid and dipyrone (metamizole) are sometimes combined in pharmaceutical dosage forms to relieve pain and fever.…”

“…Thin-layer chromatography has been widely used to determine ascorbic acid concentrations in foodstuffs (41)(42)(43), pharmaceutical preparations (43)(44)(45), and biological materials (43,46,47). Mushrooms contain reducing substances with chemical properties similar to those of ascorbic acid, and osazones were formed from the reducing substances in 19 kinds of edible mushrooms (42).…”

“…Further methods can be found using LC-MS [14,15], capillary electrophoresis [16][17][18][19][20][21] and micellar electrokinetic chromatography [17,20,22]. Other methods like fluorimetric determinations [23], sequential injection analysis [24] and thin layer chromatography [25] are also available.…”
