The methods of analysis of sulfonamides (SFAs) using CE are reviewed. Sulfonamides were the first antimicrobial group of drugs used in medical treatment. These compounds are still used today in medicine and widely are employed in veterinary medicine, also for their growth promoter effects. Improved methods of analysis of sulfonamides are a constant challenge for researchers. CE is a new trend in the fields of pharmaceutical and food analyses. Several methods for the determination of SFAs by CE have been published in recent years, and the present review considers applications in quality control of pharmaceutical dosage forms, food analysis, determinations in serum, and other biological fluids as well as in electrophoresis experiments which examine the behavior of this class of compounds for theoretical studies of the technique. This review covers studies ranging from the pioneering works on sulfonamide analyses using classical electrophoresis to the more recent CE methods coupled to tandem mass spectrometers. The sections are divided following the EC modes like CZE, MEKC, and hyphenated methods (CE-MS, CE-MS/MS). A brief compilation of theoretical findings of sulfonamides electrophoretic behavior is also included. Parameters such as recoveries, LOD and LOQ, among others, are examined, covering works published until August 2008. This review can contribute to further research aimed to improve the analysis of SFAs by CE.

Abstract. Twenty-two sulphonamides and three dihydrofolate reductase inhibitors (DHFR) were included in this study. At pH 7.5, eighteen drugs were resolved in 22 min. Above pH 7.5 the DHFR are not resolved and the pH range of 7-7.5 was found to be optimal. Around pH 7, where the degree of ionization of the SFA is highly variable, pK, and migration time are strongly correlated. Increasing phosphate buffer concentration from 25-100 mM generally improves resolution but increases analysis times. A good correlation between analyte mobility and the ratio of net charge to molecular weight indicates that this ratio may be used as a quick guide to the possibility of separation of related compounds. Our secondary objective was to compare the lead time and effectiveness of CZE with LC. It is clear that method development is much more rapid in CZE but with a sufficiently diverse group of compounds, CZE can be incapable of providing a full analysis without gradient control. Nonionized compounds are not separated at one extreme, and excessively mobile species may not emerge in a reasonable time, or, may migrate to the wrong electrode.

Capillary electrophoresis has been applied the analyses of many clinical drugs due to its rapid, high-resolution separation. In this study, electrokinetic chromatography involving the combination of SDS and dextran sulfate, which are synthetic polymers, was examined in order to obtain high resolution. Use of 2% dextran sulfate (10,000 molecular weight), 20 mm SDS running buffer containing boric acid solution (pH 9.2) and a silica capillary (inner diameter of 75 micro m, effective length of 50 cm, 57 cm overall length) afforded separation of 10 kinds of benzodiazepines. The detection limit was 0.2 micro g/mL; additionally, reproducibilities were de fi ned as the peak height and migration time. The average peak height was 5.92% (2.46-17.61), whereas the average migration time was 0.44% (0.18-0.76; n = 5). This separations system can be applied to the analysis and measurement of other pharmaceuticals as well.
