“…Since CE-ESI-MS was first applied for analysis of DNA adducts in 1994 by Janning et al [75], numerous studies have been conducted to detect modified oligonucleotides and modified nucleotides. The researches that have performed on determination of modified oligonucleotides included styrene oxide-modified oligonucleotides [75,82], alkylated oligonucleotides and oligonucleotides containing halogenated nucleobases [79], N-(acetylamino) fluorine-modified oligonucleotides [83], and unmodified oligonucleotides [77]. The modified nucleotides that have been quantitatively studied included N 7 , O-6, N 2 -modified guanosine monophosphate [82],…”

“…After reaction of styrene oxide with DNA and the digestion into oligonucleotides using the above described procedure, styrene oxide mono-adducts were detected in di-, tri-, and tetranucleotides, whereas pentanucleotides exhibit monoand discernible amounts of di-adducts. Hexanucleotides were generally modified twice [77]. Results also indicated that styrene oxide did not exhibit sequence specificity, but showed strong preference for purines versus pyrimidines in in vitro experiments.…”

“…The presence of covalently modified oligonucleotide analytes was verified by full scan MS analysis in the negative ion mode, but the alkylation site of styrene oxide at the nucleobase could not be deduced this way. In subsequent work, on-line sample stacking was employed to enhance sensitivity of the assay [77,78]. Using reaction products of styrene oxide with mononucleotides, fragments were obtained under CID giving some indication about the reaction sites in the molecules.…”

“…Therefore, molecules with a wide range of molecular weights, solubilities, and polarities, especially nonvolatile, thermally labile molecules, can be analyzed directly without derivatization. With such HPLC/ ESI/MS technology, DNA adducts may be analyzed as substituted bases (58,59), nucleosides (58, 60 -62), nucleotides (63,64), or oligonucleotides (65)(66)(67). In this report, we have demonstrated the application of LC/ MS/SIM and LC/MS/MS/SIM with ESI for the detection and quantitative analysis of the DNA adduct GEG formed in rat and fish liver tissue samples after in vivo exposure to 1,2-dihaloethanes.…”
