The epiglottis plays an important role in deglutition in humans. The present study investigated age-related changes in the epiglottis using macroscopic and microscopic measurements. Epiglottic specimens from 281 Japanese adult cadavers (177 males, 104 females) were obtained. Specimens were divided into three groups according to age: group I: 20–39 years old (32 males, 26 females), group II: 50–69 years old (82 males, 36 females), and group III: 80–98 years old (63 males, 42 females). Width, height, and thickness were measured macroscopically. To evaluate the degree of calcium deposition, the calcium volume in digitalized von Kossa-stained sections was assessed using a quantitative analysis. An elemental analysis of the area detected with von Kossa staining was done using energy-dispersive X-ray fluorescence spectrometer (EDX). Measurements of the thickness and cell density in the superficial and deep layers of epiglottic cartilage were performed in horizontal histological sections. No significant differences in macroscopic width or height were found across the age groups in either sex. A series of three measurements in males was significantly larger than in females (p < 0.05). The volume of the calcium deposit area was greater in males than in females (p < 0.05) and was significantly increased in group III in males (p < 0.05). The lower level of the epiglottic cartilage showed a greater calcium deposit area than the upper level. In the scanning image by line and surface analysis using EDX, the calcium deposit areas detected with von Kossa staining indicated a close association of calcium and phosphorus ions. The mean Ca/P molar ratio in the calcium deposit area was 1.32 ± 0.12. Microscopic cartilage thickness increased significantly with age (p < 0.05), and was greater in males than in females (p < 0.05). Cartilage cell density in the superficial cartilage layer was higher than in the deep layer and was decreased in group III (p < 0.05). Cartilage cell density was lower in males compared to females. Diameter of chondrocytes significantly increased in group III (p < 0.05) and was larger in males than in females in group III (p < 0.05). Epiglottic cartilage exhibited marked sex-related differences and progression of calcification with age. Calcification of epiglottic cartilage in elderly individuals may affect movement patterns in deglutition.

Age at death estimation from bone histology in Malaysian males

Estimation of age from microscopic examination of human bone utilizes bone remodeling. This allows regression equation to be determined in a specific population based on the variation in osteon turnover in different population. The aim of this study was to provide age estimation for Malaysian males. Ground undecalcified cross sections were prepared from long limb bones of 50 deceased males aged between 21 and 78 years. Ten microstructural parameters were measured and subjected to multivariate regression analysis. Results showed that osteon count had the highest correlation with age (R ¼ 0.43), and age was estimated to be within 10.94 years of the true value in 98% of males. Cross-validation of the equation on 50 individuals showed close correspondence of true ages with estimated ages. Further studies are needed to validate and expand these results.

RESUMENSe presenta un estudio sobre una muestra de 123 radiografías digitales de tórax de sujetos entre 5 y 75 años de edad en las que se ha analizado el estado de osificación del cartílago costal de la primera costilla. En este estudio el estadio 0 de osificación suele asociarse con edades entre 5 y 19 años. Los estadios de osificación 2 y 3 se han asociado con edades mínimas de 24 años de edad. Autores previos (Michelson, 1934) ya habían indicado que la edad mínima a la que los sujetos estudiados presentaban un estadio 3 de osificación era de 21 años de edad. Se sugiere la posibilidad de integrar el estudio de la osificación de la primera costilla entre los datos diagnósticos manejados para la estimación forense de edad en sujetos vivos basada en las recomendaciones del AGFAD del año 2000.Palabras clave: Primera costilla; osificación; cartílago costal; edad; estimación forense de la edad. Our sample is constituted by 123 digital x-rays studies had made up our sample. Minimum subject age has been 5 years old and maximum age 75 years old. Stage 0 has been usually found out in subjects between 5 and 19 years of age. Stages 2 and 3 had been found out in subjects at a minimum age of 24 years old. Previous researches had pointed out that stage 3 could be observed in subjects at an age of at least 21 years of age. We suggest that the analysis of the ossification degree of the first rib cartilage could be useful as a complementary method for the forensic estimation of age based on radiographic methods.
