Determination of Regional Intestinal Permeability of Diclofenac and Metoprolol Using a Newly-Developed and Validated High Performance Liquid Chromatographic Method

Abstract: 0.319 ± 0.184, 0.639 ± 0.241 and 0.84 3± 0.517 x 10 -4 cm/sec, respectively, for metoprolol tartrate while the corresponding permeability values were 1.585 ± 0.729, 1.154 ± 0.433 and 1.775 ± 1.576 x 10 -4

“…Also, the paper is presented in the chromatogram obtained by analysis of 100% standard solution under optimized conditions in Figure 1. In the literature, there are few HPLC methods in which PR [19] or both [21,22] as markers for SPIP studies. The optimized method appears to be more successful when evaluated in terms of chromatographic separation of previous MPT and PR analytes [21,22].…”

The optimized method appears to be more successful when evaluated in terms of chromatographic separation of previous MPT and PR analytes [21,22]. Since both PR and MPT are highly polar, the organic components of the mobile phase, methanol and C18 in the stationary phase, and phosphate buffer were preferred.…”

“…Animals were allocated one of the four groups given in Table 2. In situ single pass intestinal perfusion test procedure used in this study was a modification of the previously published reports [21][22][23][24][25]. Under anaesthetizia (60 mg/kg pentobarbital, intraperitoneal), the rats in a supine position were placed on an operation table at 37°C.…”

“…Phenol red was used as a zero permeability marker in all groups for correction of permeability coefficients of tested drugs for water transport [24]. Metoprolol tartrate was not used in group 2 and group 3 because NBV Peff value in these groups (group 2 and group 3) was compared with the NBV Peff values in groups 1 and 4.…”
