Perfluoroalkyl and polyfluoroalkyl substances (PFAS) are of increased concern because of their bioaccumulation in humans and the biota, the health risk they pose to humans and other animals, and their persistence in the environment. In the present study, the occurrence of PFAS in selected tissues from pike (Esox lucius L.) and roach (Rutilus rutilus L.) collected from two lakes in the Masurian Lake District (Poland) in eastern Europe was addressed. Ten PFAS were analyzed in the tissue of the brain, liver, kidneys, gonads, and muscles by micro-high-performance liquid chromatography coupled with tandem mass spectrometry. Only perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorononanoic acid, and perfluorodecanoic acid were detected above the limit of quantification. Concentrations in pike and roach from Ełckie Lake and Han ´cza Lake were estimated to be 14/27 and 4.4/3.2 ng/g wet weight, respectively. The contributions of PFOS and PFOA to the sum of PFAS calculated for particular tissues for each fish species were higher than those of the other analyzed compounds: PFOS was found to be predominant in fish from Ełckie Lake, whereas PFOA predominated in fish from Han ´cza Lake. It was noted that PFAS concentrations in tissues declined in the following order: kidney > gonads ≈ brain > liver > muscle. The sum of the greatest estimated PFAS concentration was 9.7 ng/g wet weight in kidneys of pike collected from Han ´cza Lake. No correlation was noted between PFAS concentration and fish size. The information provided in our study gives a better understanding of the potential dependencies in PFAS distribution and accumulation in biota.

Development of a sample preparation method for acrylamide determination in cocoa via silylation

This paper reports rapid and simple sample preparation method development for acrylamide (AA) determination in cocoa powder through conversion to N, O-bis(trimethylsilyl)acrylamide. AA level was quantified in the relation to a acrylamide-d 3 internal standard using gas chromatography coupled with ion-trap mass spectrometry in the single ion monitoring mode (GC-SIM-MS).Analytical procedure consisted of the extraction with organic solvent (acetonitrile with/without hexane addition), extract purification by dispersive solid-phase extraction (d-SPE) using various sorbents (SAX, C18, ENV, Florisil) apart from PSA sorbent, followed by derivatisation with N,OBis (trimethylsilyl) trifluoroacetamide (BSTFA) and finally with chromatographic analysis (GC-SIM-MS) using deuterated internal standard. Nine variants of the method were tested. The usefulness of the method has been verified based on the recovery ratio of acrylamide. The obtained results showed that the best recovery of 98.2% with RSD lower than 10% was obtained using a combination of sorbents PSA and C 18 with the addition of hexane in the initial stage of the extraction. The developed method has been successfully applied to determination of acrylamide in 9 cocoa powder and 30 chocolate (dark, milk and white) samples purchased at Polish supermarket.The results of AA content ranged from 54.2 to 168.0 µg kg -1 and from 25.5 to 256.1 µg kg -1 for cocoa powder and chocolate, respectively. The obtained results were in good agreement with literature reports.

Perfluorinated alkylated substances (PFASs) are today considered persistent, toxic, and bioaccumulative contaminants. Perfluorooctansulfonate (PFOS) and perfluorooctanoic acid (PFOA) are currently listed as priority substances under the UNEP global convention for the regulation of POPs. A previous study reported higher levels of PFASs in pine needles near ski areas. Their application as stain repellents in modern outdoor clothes and in ski waxes is assumed to be a potential source. Pine trees (Pinus mugo in Slovakia and Pinus sylvestris in Norway) were chosen for sampling in ski resorts. Relative distributions, overall concentrations, trend estimates, elevation patterns, and distance from primary sources were assessed. PFOA was the predominant PFAS constituent in pine needles from Slovakia (8-93%). In Norway, the most-abundant PFAS was perfluorobutanoic acid (PFBA: 3-66%). A difference in product composition (particularly in ski waxes) and differences in Norwegian and Slovakian regulations are considered to be the primary reason for these differences. Open application of PFOA in industry and products has been banned in Norway since 2011. The replacement of PFOA with short-chain substitutes is thus considered the reason for the observed pattern differences in the analyzed pine needles. Regular monitoring and screening programs are recommended.
