Determination of Glucan Contents in the Fruiting Bodies and Mycelia of Lentinula edodes Cultivars

Abstract: Shiitake mushrooms (Lentinula edodes) containing β-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The objective of this study was to determine the β-glucan content in different sections of the fruiting bodies and mycelia of ten shiitake mushroom cultivars. The measured β-glucan content ranged from 20.06 ± 1.76% to 44.21 ± 0.13% in the pileus sections, and from 29.74 ± 1.40% to 56.47 ± 4.72% in the stipe sections. The result… Show more

“…The repeating basic units of β-glucan polysaccharides derived from edible shiitake ( Lentinus edodes ) mushrooms consist of five linearly linked (1→3)-β-glucose residues and two 1-6-β-glucopyranoside branches [ 30 ]. To determine the β-glucan content of shiitake mushrooms, these authors used a mushroom specific β-glucan kit on different sections of fruiting bodies and mycelia of ten shiitake mushroom cultivars.…”

“…On the other hand, NEH-glucose might be released mostly from the complex fungal cell wall polysaccharides comprised of branched β(1,3)/(1,6) glucan linked to chitin. The complex polymer requires pretreatment with concentrated acid to achieve complete hydrolysis to compositional monomers (Dallies et al 1998;Francois 2006;Won et al 2014). Previously, fresh ECM fungal sclerotia were not observed to incorporate roots or root hairs, although they were formed very close to pine roots (Grenville et al 1985).…”

“…After slight modification by Manzi and Pizzoferrato [ 60 ], it was used for the determination of β-glucans in fungi. In later years, the determination of β-glucan content by enzymatic method in fungi was carried out by Bak et al [ 61 ], Cha et al [ 53 ], and Kang et al [ 54 ]. This enzymatic method with modifications is currently recommended by the American Association for Clinical Chemistry (AACC).…”
