Determination of Gabapentin in Serum Using Solid-Phase Extraction and Gas-Liquid Chromatography

Abstract: A gas-liquid chromatographic method for the determination of gabapentin (Neurontin) is described. The method involves extracting 0.5 mL of acidified sample by C18 solid-phase column, derivatization with MTBSTFA plus 1% tBDMCS, and analysis on an HP-1 column with a flame-ionization detector. Quantitation was performed with peak-height ratios of gabapentin to a gabapentin analogue [(1-aminomethyl-1-cycloheptyl) acetic acid] as the internal standard. The assay had a limit of detection of 0.2 mg/L and a linear ran… Show more

“…Gas chromatographic methods require derivatization. GC with flame ionization detector is used for the determination of gabapentin [11] and GC/MS methods are reported for the determination of gabapentin and vigabatrin [12,13]. Numerous HPLC methods have been published about the determination of gabapentin, pregabalin or vigabatrin in biological fluids using different derivatizing reagents [14 -16].…”

“…The method most commonly used for GBP quantification in blood or serum is reversed-phase HPLC with derivatization and fluorometric detection [146][147][148]. GC methods have also been reported [149], and more recently, more sensitive and validated LC procedures with MS or MS/MS methods for both clinical and experimental monitoring of GBP in human and animal have been also been described [149][150][151][152][153].…”

“…Several types of analysis have been performed, including spectrofluorimetry (15)(16)(17) and HPLC with a variety of detection methods, including mass spectrometry and capillary electrophoresis, but most frequently, fluorescent derivatization (18 -23 ). Gas chromatography (GC) has also been used with detection by mass spectrometry (MS) and flame ionization (24,25 ). Here we describe a rapid and cost-effective isocratic liquid chromatography method that uses commonly available ultraviolet detection for gabapentin quantification without the need for solid-phase extraction.…”
