Determination of Corticosterone Concentration in Plasma of Turkeys Using Radioimmunoassay

Abstract: A method of determining corticosterone concentration in turkey plasma was developed using radioimmunoassay. Compared to fluorometric analysis and competitive protein-binding radioassay, this method had the following advantages: 1) results were consistent, 2) the method was simple and rapid to perform, 3) only 20 lambda of plasma was required, 4) all procedures were performed using the same tube except for counting, and 5) unknown and standard samples were treated identically. The coefficient of variation with … Show more

“…Cortisol was measured as total cortisol based on Simensen et al (1978), with the following modifications for polar bear plasma: 10-µl plasma samples were heated in 500 µl phosphate-buffered saline (PBS) with 0.1% gelatin for 20 min to remove possible inhibiting effects from plasma proteins. Validation showed that serial dilution of heated polar bear plasma with high cortisol concentration produced a dose-response curve parallel to the standard curve.…”

“…The plasma cortisol concentration was quantified using a radioimmunoassay (Simensen et al, 1978;Olsen et al, 1992). Liquid scintillation (Packard Tricarb 1500) was used to determine sample radioactivity.…”

“…In ducks, although the influence of some stresses, including adrenocorticotrophic hormone (ACTH; Macchi, Phillips & Brown, 1967;, cold (Boissin, 1967;Ensor, Phillips & O'Halloran, 1976;Holmes & Slikker, 1976;Holmes, 1978) and osmotic stresses (Macchi et al 1967;Allen, Abel & Takemoto, 1975) in enhancing secretion of corticosterone has been determined, the fluorometric and competitive protein-binding assays used in these studies are known to over-estimate levels of corticosterone in plasma (Culbert & Wells, 1975;Holmes & Phillips, 1976), as well as giving inconsistent results for birds (Simensen, Olson, Vanjonack, Johnson & Ryan, 1978). In view of the concern about stress in the poultry industry (Ministry of Agriculture, Fisheries and Food, 1965;Sorensen, 1978), the possible influence of stress on the secretion of corticosterone in the duck has been further investigated in the present study by using a sensitive radioimmunoassay for corticosterone.…”
