Determination of Antioxidant Components and Activity of Tamarix ramosissima Comparative with Vaccinium myrtillus on Streptozotocin-diabetic Mice

Ocimum sanctum L. is widely used as traditional remedy to manage hyperglycemia. This study was aimed to evaluate the effect of aqueous-methanolic extract of O. sanctum leaves (OSE) on the anti-diabetic activity of gliclazide in alloxan-induced diabetes in Wistar rats. Diabetes was induced by intraperitoneally injecting alloxan (120 mg/kg b.w.) in rats. Treatments including OSE (100 mg/kg b.w.), gliclazide (100 mg/kg b.w.), and in combination were given daily to diabetic rats till the 21st day of study. Body weight and fasting blood glucose levels were determined at regular intervals, while blood and organ samples were taken at the end of the study for biochemical and histopathological studies. Results showed that treatments exhibited anti-hyperglycemic activity through significantly (p [ 0.05) restoring body weight, fasting blood glucose level, and serum levels of glucose, insulin and HbA1c. The anti-lipidemic activity was noticed as total cholesterol (TC), triglyceride (TG), high-/low density-lipoproteins (HDL-C, LDL-C) levels were restored in treated diabetic rats. Ameliorative effects of treatments were observed as significant (p [ 0.05) reduction in serum levels of liver function biomarkers (alanine aminotransferase; ALT, aspartate aminotransferase; AST, alkaline phosphatase; ALP and bilirubin; BIL) and restoration of oxidative stress biomarkers (catalase; CAT, superoxide dismutase; SOD and malondialdehyde; MDA) in liver tissue. Histopathological findings supported these results as an increase in pancreatic islets size and protective effects on liver tissue was observed in diabetic rats treated with gliclazide and OSE alone and their combination. Conclusively, the combination of OSE and gliclazide produced a synergistic anti-diabetic effect as compared to that of alone treatment.

Determination of the Polyphenolic Content of Dorycnium herbaceum Plant Extract and Testing in vivo Antioxidant Effect, Comparative with Bilberry

Dorycnium herbaceum (DH) is a perennial herb or small shrub 10-80 cm high, which can be found in Central and Southeastern Europe. It is a little studied plant, which is known to have antibacterial and antifungal effect. In the present study we determined by spectrophotometric methods the total polyphenolic, flavonoidic, carotenic and chlorophyllic content of DH species, comparative to the bilberry. We also monitored the status of antioxidant enzymes using mice with oxidative stress induced by streptozotocinic diabet. For the induction of diabetes we used Swiss albino mice to which we injected streptozotocin intraperitoneally at a single dose of 180 mg / kg b.w.. The results showed that both chemical composition tinctures contain appreciable amounts of polyphenols, the highest content starring DH tincture species, but bilberry fruits have higher content of flavonoids, carotene and chlorophyll. Both plant extracts regulate the status of antioxidant enzymes, DH having superior efficacy.

The last century was marked by efforts to search for natural compounds with antibacterial therapeutic properties, due to the gradual reduction of the number of effective allopathic antibiotics and on the toxic effects of antibiotic residues in animal products. Medicinal plants are considered new resources for producing agents that could act as alternatives to antibiotics in the treatment of antibiotic-resistant bacteria. We determined the polyphenolic composition of the tested plant extract by HPLC technique. The aim of this study was to evaluate the antibacterial activity of Vaccinum myrtillus folium (VM-f) ethanolic extract, using the diffusion method on nutritive agar (Kirby-Bauer). We compared the experimental results with the theoretical antibacterial activity calculated with AUTODOCK 4.2.
