Determination of amobarbital and secobarbital in plasma samples using micellar liquid chromatography

Abstract: A new liquid chromatographic procedure for the determination of amobarbital and secobarbital in plasma samples is proposed. The method uses a Spherisorb octadecylsilane ODS-2 C(18) analytical column, a guard column of similar characteristics and 0.04 M CTAB solution buffered at pH 7.5 containing 3% 1-propanol as micellar mobile phase. The UV detection was carried out at 250 nm. Butabarbital was used as internal standard. Plasma samples preparation only required adequate dilution with the mobile phase before in… Show more

“…With their BMC retention factors, these authors obtained good correlations to different bioactivity data of several groups of xenobiotics. [265][266][267] Whether the Spanish group found a universal biopartition modeling system remains to be independently confirmed. Nonetheless, regarding the behavior in conventional reversedphase HPLC, MLC has several advantages as the method of assessment of lipophilicity of xenobiotics.…”

“…Quantitative methods for the analysis of barbiturates in biological samples have been described in the scientific literature. Techniques such as micellar liquid chromatography , liquid chromatography‐MS , and GC‐MS have been used . Conventional sample preparation techniques such as liquid–liquid extraction (LLE) and SPE have been used to extract barbiturates from biological specimens especially for gas chromatographic analysis .…”

“…The use of special coupled-columns (RAM-columns) [11][12][13] that allow the direct injection of plasma samples has also been proposed. Different analytical procedures that use micellar mobile phases for determining different kind of drugs in biological samples have been reported [16][17][18][19][20][21]. Micellar liquid chromatography (MLC) is an alternative to RP-HPLC that uses aqueous solutions of surfactants above the critical micellar concentration as mobile phases [14].…”
