Detection of Serum Antibodies to S‐Antigen by Surface Plasmon Resonance (SPR)

Abstract: Serum autoantibodies to visual arrestin, also termed S-antigen, have been shown to accompany several autoimmune-related diseases. However, they were also detected in sera of healthy individuals; there is lack of a sensitive and fast method for evaluation of putative differences between those two groups of antibodies. We show that, using biosensor technology based on surface plasmon resonance (SPR), it was possible to characterize real-time interactions of immune sera with immobilized arrestin. Binding characte… Show more

“…The technology's breadth of applications is further illustrated by the range of articles investigating oligonucleotides , small molecules , carbohydrates , and extracellular matrix components . In addition, the number of articles describing biosensor work in clinical support [1008][1009][1010][1011][1012][1013][1014][1015][1016][1017][1018][1019][1020][1021][1022][1023][1024], food/agricultural/veterinary/environmental sciences [1025][1026][1027][1028][1029][1030][1031][1032][1033][1034][1035][1036][1037][1038], and membrane/virus/cell characterization [1039][1040][1041][1042][1043][1044][1045][1046][1047][1048][1049][1050] demonstrate Biacore's emerging contributions in these fields. Biacore's more unusual applications, including the characterization of fibrils and the analysis of crude analytes are summarized in References 1051-1066.…”

“…The SPR has been used extensively to study antigen/antibody interactions and measure their binding kinetics 50,51 . In majority of diagnostic interventions, this technique has been utilized to detect presence of antibodies in serum using an immobilized antigen ligand on the chip 52 . SPR assays has been reported for detecting protein biomarkers in human sera in cases of pancreatic cancer 46 , ovarian cancer 49 and coronary artery disease 24 .…”

“…Very few studies have measured biomarkers in blood samples using SPR biosensors. [15][16][17][18][19] An in situ filter consisting of micro slit arrays was fabricated on the gold surface of an SPR device for on-chip blood cell filtration and biomarker detection in one step. 20 Using this setup, detection of IgG from a whole blood sample in 100 s of application was demonstrated.…”
