Abstract. Hanafiah M, Helmi TZ, Sutriana A, Bahi M. 2021. Morphology and molecular identification of Eurytrema spp. worm in Aceh cattle, Indonesia. Biodiversitas 22: 5654-5661. The objective of this research was to determine the species of Eurytrema spp. in Aceh cattle which were slaughtered in slaughterhouses located in Banda Aceh, Indonesia. Identification of Eurytrema spp worm species was performed by Semichon’s Carmine staining, SEM and molecular method. The result fromSemichon’s Carmine staining and SEM method showed that worms coded E2 and E3 were Eurytrema cladorchis indicated by clearly visible protruding cirrus while worms coded E1, E4 and E5 were E. pancreaticum indicated by the absence of protruding cirrus. Molecular characterization of Eurytrema spp.produced fragments of approximately 700 bp in length. The phylogenic tree showed that Eurytrema spp.from Aceh cattle consisted of 2 clades: clade 1 was E. pancreaticum, while clade 2 was E. cladorchis. DNA sequencing result showed that the similarity of worm samples coded 1, 4, and 5 were 98.90% similar to E. pancreaticum (KJ767631.1 and KC535543.1), and 96. 33% similar to E. cladorchis (MN566134.5 and MN566140.1), whereas the worm samples coded 2 and 3 were 97.53% similar to E. pancreaticum (KY490000.1 until KY490004.1), and 97.55 % similar to E. cladorchis (MN566135.1 until MN566140.1). This research was able to identify E. pancreaticumand E. cladorchis in Aceh cattle slaughtered in Banda Aceh slaughter houses.

Background: Permanent staining of helminths is important for different purposes including taxonomic and prevalence studies as well as teaching. Carmine is one of the most commonly used dyes; however, variable factors affect the quality of the staining outcomes. Therefore, it is essential to find an alternative stain that is efficient, accurate, and less complex. Objective: The aim of the present study is to evaluate the quality of a combined lactophenol methylene blue-blue glycerol jelly (LMB-BGJ) technique as a potential alternative for staining of helminths compared with carmine-based staining and glycerol jelly mounting techniques. Material and Methods: S. mansoni and T. spiralis were obtained from experimentally infected animals. A new staining technique (LMB-BGJ) was used for both parasites in comparison with carmine stain in case of S. mansoni and glycerol jelly mounting in case of T. spiralis. Comparison was conducted using examination under light microscope and a mean score was given for the structural brightness, and extent of staining of tegument/cuticle, suckers, intestine, as well as reproductive systems. Results: S. mansoni worms prepared with both carmine staining and LMB-BGJ showed moderate staining quality of tegument, suckers, intestine and reproductive organs with a mean total score of 3.3/4 and 3.35/4, respectively. Adult T. spiralis mounted with glycerol jelly only showed a mean total score of 3.95/4 for the cuticle, esophagus, and intestine, while the LMB-BGJ showed a staining quality with a mean total score of 3.85/4. Conclusion: From a practical point of view, LMB-BGJ is a simple and rapid technique that needs less experience for the preparation of semi-permanent slides with satisfactory staining and mounting quality for both trematodes and small nematodes.

Kerbau lumpur (Bubalus bubalis) merupakan ternak yang memegang peranan dalam kehidupan ekonomi dan sosial masyarakat Kabupaten Sumba Timur. Salah satu penyakit yang hingga kini masih menjadi masalah kesehatan pada ternak, termasuk kerbau, adalah infeksi Trematoda. Penelitian ini bertujuan mengidentifikasi jenis cacing Trematoda yang menginfeksi serta mengukur prevalensi dan intensitas infeksinya. Metode filtrasi bertingkat digunakan untuk mendeteksi keberadaan Trematoda pada penelitian ini. Hasil pemeriksaan 105 sampel tinja kerbau asal Kabupaten Sumba Timur menunjukkan sebanyak 17/105 (16,19%) kerbau mengalami Trematodosis yang disebabkan oleh infeksi tunggal Fasciola gigantica (0,95%; rataan telur tiap gram tinja [TTGT] 1,00) dan Paramphistomum sp. (14,29%; rataan TTGT 2,26), serta infeksi campuran (koinfeksi) kedua Trematoda tersebut (0,95%; rataan TTGT 2,45). Prevalensi infeksi Trematoda kerbau betina (17,33%) ditemukan jauh lebih tinggi dibandingkan dengan kerbau jantan (13,33%). Umur dan jenis kelamin diketahui tidak berasosiasi nyata (p > 0,05) dengan kejadian dalam penelitian ini. Intensitas infeksi Trematoda pada penelitian ini dikategorikan ringan, dengan rataan geometrik 2,18 TTGT. Temuan ini menunjukkan bahwa kerbau lumpur di Kabupaten Sumba Timur terinfeksi oleh cacing Trematoda, sehingga diperlukan upaya pengendalian untuk mencegah kerugian akibat infeksi tersebut.
