Detection of Microsporidia Infecting Beet Webworm Loxostege Sticticalis (Pyraloidea: Crambidae) in European Part of Russia in 2006–2008

Abstract: The beet webworm Loxostege sticticalis (L.) is a major insect pest that causes serious damage of agricultural crops in Russia, China and adjacent countries. Microsporidia are obligate intracellular parasites that negatively affect population density of many insect hosts including Lepidoptera. In particular, infection with microsporidia is an important mortality factor for L. sticticalis. Special methodology for the identification of microsporidia associated with terrestrial insects is required. In the present … Show more

“…Alive and perished larvae were dissected and inner tissues were processed for molecular genetic analysis. Genomic DNA was extracted as described previously [12]. Degenerate primers BgDNVfor3 (5′-GWMAGGGWGACGAGCAACGTTC-3′) and BgDNVrev3 (5′-RTTTCCCTWGGTTCRTARTTWGGTTC-3′) were designed to span a fragment, ∼800 bp long, corresponding to the non-structural protein within the open reading frame 3 (ORF3) closer to its 3′-end, in the genomes of two densoviruses presented by Genbank accession #AY189948 and #JQ320376.…”

“…For DNA analysis, genomic DNA was extracted routinely (Malysh et al., 2019) from spore samples, infected larvae, and experimental egg batches, followed by PCR and amplicon sequencing with 18f:1047r flanking a ca. 850 bp long region of microsporidia small subunit ribosomal RNA (SSU rRNA) gene (Weiss & Vossbrinck, 1999) and NbHKfor1:NpyrHKrev1 primers flanking a 709 bp long region of N. pyrausta hexokinase (Tokarev et al., 2019).…”
