Aims: To investigate the fate of a streptomycin–rifampicin‐resistant variant of Lactobacillus gasseri SBT2055 (LG2055SR) and the influence of its oral administration on the composition and metabolism of the intestinal microflora. Methods and Results: Intestinal passage of LG2055SR was monitored by a combination of selection with antibiotics and identification by a randomly amplified polymorphic DNA (RAPD)‐PCR method. Composition of intestinal microflora was analysed by the method developed by Mitsuoka et al. (1965, 1974). Establishment of orally‐administered LG2055SR in the human intestine was confirmed in this study. LG2055SR ingestion specifically lowered faecal populations of Staphylococcus and faecal contents of p‐cresol. Conclusions: LG2055SR and its parent strain, LG2055, are considered to be appropriate candidates for probiotics. Significance and Impact of the Study: It is clarified that LG2055SR has the ability to establish in the human gastrointestinal tract and alters the composition and metabolism of the intestinal microflora and physical characteristics of faeces.

Probiotics are initially defined as food supplements containing live microorganisms which beneficially affect the host by improving intestinal microbial balance. Although the definition has been expanded by other researchers from time to time, in this view, lactic acid bacteria (LAB) are unalterably considered to be the main components of probiotics as they fulfill, in the best course, the requirements targeted by the basic idea of these expanded definitions. It has been believed that periodical supplementation of special cultures of probiotic LAB is helpful to keep the person who consumes it in good health. Recently, the ingestion of LAB as a probiotic has drawn much interest all over the world because of growing health consciousness and concern. A typical example is seen in Japanese society. Well-being is one of the most important matters of concern for Japanese because of the rapid aging of the society. The beneficial image of probiotics has led to an increase in the consumption of fermented milk products in Japan as well as in other countries. Bifidobacteria are often used as dietary supplements or as starter cultures in the production of fermented milk products used as health foods. I have developed the probiotic strain Bifidobacterium longum SBT2928 (BL2928) over a decade for this purpose. This review summarizes the research done to demonstrate the biological functions of BL2928, especially regarding its effect on the host's immune system, on intestinal transit as well as the intestinal microflora composition and metabolism, and specific competitive exclusion of enterotoxigenic Escherichia coli by a novel anti-infectious factor (BIF).

Clostridium butyricum has been used as a probiotic in animals and humans for years, however, its fate in the intestine has not been clarified yet. We investigated the intestinal fate of C. butyricum using a selective medium and a monoclonal antibody after orally administering C. butyricum spores to rats. The number of C. butyricum, both viable and dead cells, in the intestinal contents were counted by enzyme-linked immunosorbent assay (ELISA) at various times after a single oral administration. The total viable number of C. butyricum was counted using a selective medium, and viable resting spores were selectively detected by treating the samples with ethanol. To investigate the intraluminal localization of the C. butyricum cells, frozen intestinal tracts were imprinted onto slides and stained with immunogold-silver. Total viable spores exceeded the number of viable resting spores by more than 10-fold from the proximal to middle of the small intestine 30 min after administration. Vegetative cells of C. butyricum were first detected in the dis-tal small intestine after 2 hr, and vegetative growth was observed from the cecum to the colon 5 hr after administration. Dead vegetative cells were detected 9 hr after administration, and C. butyricum cells were not detected in the intestine after 3 days. The C. butyricum cells in the intestinal imprints were stained specifically by immunogold-silver staining, and proliferative cells were observed in the cecum after 3 hr. These results suggest that the administered C. butyricum germinated in the upper small intestine, grew mainly from the distal small intestine to the colon and were excreted from the rat intestine within 3 days.
