Detection of Human Cytomegalovirus in patients with Colorectal Cancer by Nested-PCR

Abstract: Background: The association of colorectal cancer with human cytomegalovirus (HCMV) is a controversial issue in cancer research. This study aimed to identify the HCMV virus in colorectal cancer tissues and to investigate the association of HCMV with colorectal cancer. In this study, 50 cancer tissue samples and 50 samples without colon cancer were studied in order to identify the HCMV virus through nested-polymerase chain reaction. The cases of adenocarcinoma tissues were in a moderately differentiated stage, a… Show more

“…After an initial denaturation phase at 95°C for three minutes, 45 cycles programmed were along these lines: denaturation phase at 95°C for 35 seconds, annealing phase at 55°C for 45 seconds, primer extension at 72°C for 40 seconds and final extension phase at 72°C for five minutes. PCR products were determined by visualization of amplicons on 2% agarose gels stained with Safe Stain (22).…”

“…The second-round PCR amplification was performed via 12.5μl of the Amplicon master mix, 0.5μl of forward and reverse primers and 5μl of the firstround PCR products as templates in 25μL reactions (22). The sequences of the inside primer pair were: 5'GCCCGCCGCGGCAGCACCTGGCT-3' and 5'GTAAACCACATCACCCGTGGA-3' (23).…”

“…Human cytomegalovirus (HCMV) has been detected in glioblastoma multiforme 14,15 , medulloblastoma 16 , neuroblastoma 17 , colon cancer 18,19 , and prostate cancer 20 , as well as in primary BC and sentinel lymph nodes and distant metastases from BC patients [21][22][23] . In general, increased viral activity is associated with worse prognosis.…”

“…Some earlier studies reported negative detection of HCMV in the tumour tissue of CRC (Akintola-Ogunremi et al, 2005;Boguszakova et al, 1988;Brichacek et al, 1980). However, more and more recent data have indicated that HCMV preferentially infects the tumour tissue of CRC (Cai et al, 2016;Dimberg et al, 2013;Li et al, 2015;Salyakina & Tsinoremas, 2013;Sole et al, 2015;Tafvizi & Fard, 2014;Taher et al, 2014). There is also a growing body of evidence suggesting that HCMV plays an oncomodulatory role and affects the disease course of human cancers.…”
