“…Fluorescence expression data was modeled (see below), and this model was then used to characterize the RDX detection efficacy of the riboswitch, and to provide insight into riboswitch characteristics and assay design constraints for efficient RDX detection. We generated a standard curve using purified DsRed protein (BioVision, Milpitas, CA), and measured fluorescence of the DsRed protein using a BioTek Synergy HT plate reader (BioTek Instruments, Inc., Winooski, VT) as previously described [ 18 ], with an excitation wavelength of 530 nm, an emission wavelength of 590 nm, and a PMT sensitivity rating of 35. These data establish a quantitative relationship between protein concentration and fluorescence ( S1 Fig ).…”
