Coagulase-negative staphylococci (CoNS) are a group of commensal microorganisms residing on the skin and mucous membranes of both humans and animals. Until recently, they have been regarded as non-pathogenic to livestock and companion animals, but since then, their clinical importance in veterinary medicine has increased with the discovery of their potential pathogenic roles in animals causing skin and soft tissue infections together with spontaneous abortions and mastitis. Scientific data concerning the presence of CoNS in North Cyprus are very limited. Therefore, the purpose of the study reported herein was to investigate the presence and antimicrobial resistance patterns of CoNS species isolated from various animals presented at the Veterinary Teaching Hospital in North Cyprus between July 2018 and 2019. Staphylococci were isolated from 37.0% (87/235) of the samples submitted, within which 60.9% (53/87) and 39.1% (34/87) were identified as coagulase-positive staphylococci (CoPS) and coagulase-negative staphylococci (CoNS), respectively. Among the CoNS, S. chromogenes was the most predominantly isolated species (14/34, 41.2%), followed by S. capitis (5/34, 14.7%) and S. simulans (4/34, 11.8%). Of these 34 CoNS, 24 (70.6%) and 10 (29.4%) were identified as MRCoNS and MSCoNS, respectively. The CoNS isolates showed relatively high levels of resistance towards amoxicillin/clavulanic acid (19/34, 55.9%), tetracycline (14/34, 41.2%) and penicillin (13/34, 38.2%). In conclusion, the presence of CoNS, especially MRCoNS, and the detection of multiple drug resistant (MDR) species with a high prevalence were regarded as being important since they might limit and have negative effects on the therapeutic treatment options of staphylococcal infections in animals, and might have both public and veterinary concerns.

Prevalence and Genetic Basis of Antimicrobial Resistance in Non-aureus Staphylococci Isolated from Canadian Dairy Herds

Emergence and spread of antimicrobial resistance is a major concern for the dairy industry worldwide. Objectives were to determine: (1) phenotypic and genotypic prevalence of drug-specific resistance for 25 species of non-aureus staphylococci, and (2) associations between presence of resistance determinants and antimicrobial resistance. Broth micro-dilution was used to determine resistance profiles for 1,702 isolates from 89 dairy herds. Additionally, 405 isolates were sequenced to screen for resistance determinants. Antimicrobial resistance was clearly species-dependent. Resistance to quinupristin/dalfopristin was common in Staphylococcus gallinarum (prevalence of 98%), whereas S. cohnii and S. arlettae were frequently resistant to erythromycin (prevalence of 63 and 100%, respectively). Prevalence of resistance was 10% against β-lactams and tetracyclines. In contrast, resistance to antimicrobials critically important for human medicine, namely vancomycin, fluoroquinolones, linezolid and daptomycin, was uncommon (< 1%). Genes encoding multidrug-resistance efflux pumps and resistance-associated residues in deducted amino acid sequences of the folP gene were the most frequent mechanisms of resistance, regardless of species. The estimated prevalence of the mecA gene was 17% for S. epidermidis. Several genes, including blaZ, mecA, fexA, erm, mphC, msrA, and tet were associated with drug-specific resistance, whereas other elements were not. There were specific residues in gyrB for all isolates of species intrinsically resistant to novobiocin. This study provided consensus protein sequences of key elements previously associated with resistance for 25 species of non-aureus staphylococci from dairy cattle. These results will be important for evaluating effects of interventions in antimicrobial use in Canadian dairy herds.

The discovery of MRSA encoding the mecA variant mecC from dairy cattle and humans, 1 and then subsequently from a diverse range of other host species, 2,3 has raised questions over the origin and epidemiology of this resistance determinant. In addition to Staphylococcus aureus, mecC has been described in other staphylococci, namely Staphylococcus xylosus, 4 Staphylococcus sciuri, 5 Staphylococcus stepanovicii, 6,7 Staphylococcus saprophyticus 8,9 and Staphylococcus edaphicus, 10 all of which are methicillin resistant with the exception of S. xylosus. The mecC of S. aureus is encoded within an SCCmec type XI element, carrying a class E mec gene complex associated with the genes ccrA1 and ccrB3, corresponding to a type 8 ccr gene complex. 11 This is not the situation in the other mecC-positive staphylococci reported to date. In these species, although mecC is part of a conserved class E mec gene complex inserted into the orfX region, the neighbouring sequences vary considerably between the different species with little homology to SCCmec type XI or any other known SCCmec type and do not always contain ccr gene complexes. 4-6,10 Herein we describe an S. xylosus isolate, 47-83, from bulk tank milk collected in Sussex, England in 2013, which encodes mecC within a highly conserved SCCmec type XI. The isolate was found during the screening of bulk tank milk for MRSA, as described previously. 12 It grew on MRSA Brilliance MRSA 2 agar (Oxoid, Basingstoke, UK) and was resistant to oxacillin and cefoxitin by disc diffusion. Susceptibility testing of 47-83 using the VITEK 2 card AST-P634 found it to be susceptible to chloramphenicol, ciprofloxacin, clindamycin, daptomycin, erythromycin, gentamicin, linezolid, mupirocin, rifampicin, teicoplanin, tetracycline, tigecycline, trimethoprim and vancomycin,
