Detection and typing ofBorrelia burgdorferisensu lato genospecies inIxodes persulcatusticks in West Siberia, Russia

Abstract: The prevalence of Borrelia burgdorferi sensu lato (s.l.) genospecies in West Siberia as well as in many other regions of Russia remains insufficiently investigated. In the present study a total of 151 adult female ticks Ixodes persulcatus Schulze, collected at three localities in eastern regions of West Siberia, where Lyme disease is endemic, were examined for the presence of the spirochete B. burgdorferi s.l. by polymerase chain reaction targeting the 23S^5S rRNA intergenic spacer regions. Spirochetal DNA was… Show more

“…I. minor larvae collected from the same bird were feeding for the first time, picking up Borrelia species from the same bird host. It is not unusual that the vector, the reservoir host or LB patient maintain multiple spirochete species [92-99]. However, it is difficult to explain the unprecedented level of diversity among single-bird host-associated spirochete strains, specifically those isolated from larvae, considering the recent claim that transovarial transmission of B. burgdorferi sl does not exist [82].…”

“…B. burgdorferi sensu lato 5S-23S rRNA intergenic spacer was amplified by using nested PCR with two primer pairs (Bb23SN1–Bb23SC1 and IGSb1–IGSa2) [ 24 ]. The PCR inhibition possibility was controlled by 150-bp long fragments of internal standard [ 25 ]. DNA isolated from B. burgdorferi sensu stricto B31 was used as a positive control.…”
