Designing Immunogens to Elicit Broadly Neutralizing Antibodies to the HIV-1 Envelope Glycoprotein

Abstract: To date HIV-1 vaccines have not been able to elicit potent, long lasting, and broadly neutralizing antibodies to the virus. Our knowledge of HIV envelope glycoprotein (Env) structure/function and the existence of a handful of broadly neutralizing antibodies is guiding rational immunogen design. We review here the potential targets on the HIV Env (the glycan shield, the CD4 binding site, the coreceptor binding site, Env fusion intermediates, and the membrane proximal region) and their associated rational immuno… Show more

“…Therefore, the apparent paradox of decreased neutralizing activity in the face of an overall robust humoral response can be resolved if the response is directed toward nonneutralizing epitopes. The diversion of humoral immunity away from the larger neutralizing epitope is a strategy that has been proposed for HIV and PRRSV (27,46).…”

“…Whether an outcome of viral evolution or vaccine design, these immunodominant non-neutralizing epitopes on viral RBDs can outcompete other epitopes in triggering host immune responses, so that the resulting immune responses target these non-neutralizing epitopes while neglecting neutralizing epitopes on viral RBDs (refs 7, 8, 9, 10). Rational design of viral subunit vaccines aims to focus the immune responses on neutralizing epitopes through masking or deletion of immunodominant non-neutralizing epitopes111213.…”

“…Other than complete functional trimers, natural virions may display gp120/gp41 monomers, gp120-depleted gp41 stumps, and other incomplete or false oligomeric forms that may act as immune decoys [19, 42, 43]. These alternate viral protein structures may misdirect the immune response toward non-neutralizing epitopes on the gp120 glycoprotein.…”
